The present invention provides a process for producing biologically active recombinant yeast protein disulfide isomerase, which includes the steps of: a) deleting, substituting, or adding one or more bases in a region encoding an endoplasmic reticulum retention signal in a gene encoding protein disulfide isomerase of yeast to modify the gene so as not to encode part or all of the endoplasmic reticulum retention signal; b) incorporating the modified gene into an expression vector; c) transforming host cells with the expression vector; and d) culturing the host cells transformed with the expression vector in a culture medium kept at a nearly neutral pH, thereby causing protein disulfide isomerase to be secreted in an active state outside the host cells. According to the process of the invention, recombinant yeast protein disulfide isomerase can be secreted in a large amount in a biologically active state into a culture medium, and can be collected by a simple purification method.

The present invention provides a method for producing nested deletions in vitro in desired DNA, comprising: 1) preparing a vector containing a DNA fragment in which nested deletions are to be generated and the terminal repeat of a transposon; 2) incubating the vector in vitro with transposase and a DNA replication system; 3) obtaining the vector incorporating the DNA fragment with nested deletions as a reaction product; and optionally, 4) transforming a host cell with the reaction product and growing it; and 5) recovering a vector incorporating said DNA fragment with nested deletions from the grown host cell.

An application unit moves from the rear side of the exposure unit and applies water successively to the photosensitive material. A superposing unit moves from the rear side of the application unit, and successively superposes an image-receiving material on the photosensitive material. Heat development transfer is carried out, and an image is obtained on the image-receiving material.

In the image recording apparatus, an upper cover is rotatably connected to and supported at a base stand by an intermediate hinge. Accordingly, there are a first stage opened/closed state, in which the upper cover opens and closes around a supporting shaft, and a second stage opened/closed state, in which the upper cover integrally opens and closes along with the intermediate hinge around another supporting shaft. In the second stage opened state, the inner portion of the base stand is exposed by a large amount, the distal end portion of a stage is lifted, and further, the holding of a supply magazine and a take-up magazine is released. As a result, the innermost place within the apparatus is exposed and the workability of inspection and maintenance, such as replacement of the supply magazine or the take-up magazine, improves.

The plasmid of the present invention contains Leu 2 gene (.beta.-isopropylmaleate dehydrogenase gene) from Saccharomycopsis lipolytica and an autonomously replicating sequence (hereafter referred to as ARS) and can be a shuttle vector between Escherichia coli and Saccharomycopsis lipolytica since it is produced from shuttle vector pVC 1 of Escherichia coli and Saccharomyces cerevisiae. An exogenous useful gene is inserted into the plasmid of the present invention, namely, shuttle vector to transform Escherichia coli, whereby a large quantity of plasmids are obtained. It becomes possible to produce a useful substance in Saccharomycopsis lipolytica as a host using the plasmid.

The vacuum deposition device includes a vacuum deposition chamber, a vacuum evacuator for evacuating an inside of the chamber, an evaporating unit for evaporating one or more film forming materials in the chamber, a holding unit provided above the evaporating unit for holding a substrate for the vacuum deposition, and a preventing unit for preventing evaporated particles of the one or more materials from adhering to the substrate when the vacuum deposition is carried out at a pressure of 0.05 to 10 Pa. The pretreatment method for the vacuum deposition prepares the prevention unit in the chamber and melts the one or more materials by heating in the chamber at the pressure of 0.05 to 10 Pa while preventing the evaporated particles from adhering to the substrate by using the preventing unit.
