Multiple derivatization by chemical reactions of analytes for mass spectrometry is disclosed. The derivatizations enhance the use of MS techniques for analyzing protein samples, particularly when the sequence of a polypeptide is determined by tandem MS/MS. Accurate mass analysis techniques are described for use in sequencing polypeptides, together with the use of sequencing data in protein analysis. An apparatus and method for calibrating a mass spectrometer by internally introducing calibration masses at a post-source stage of the mass spectrometer is also provided. Lock mass ions mix with the derivatized polypeptide analyte ions prior to mass analysis.

The invention provides an apparatus that produces analyte ions for detection by a detector. The apparatus includes a matrix based ion source having a nanowire target substrate for producing analyte ions, an ion transport system adjacent to the matrix based ion source for transporting analyte ions from the matrix based ion source; and an ion detector downstream from the ion transport system for detecting the analyte ions. The invention also provides a method for producing and detecting the analyte ions.

The present invention provides a method and apparatus for determining the identity of a monomeric residue of a biopolymer. The apparatus comprises a substrate having a nanopore, a potential-producing element for producing a ramped potential across electrodes adjacent to the nanopore, and a quenchable excitable moiety adjacent to the nanopore. As a biopolymer passes through the nanopore, the identity of monomeric residues of a biopolymer may be determined by detecting changes in (a) current across the electrodes and (b) a signal of the quenchable excitable molecule. The subject method and apparatus find use in determining the identity of a plurality of monomeric residues of a biopolymer, and, as such, may be employed in a variety of diagnostic and research applications.

The present invention provides an apparatus and method for use with a mass spectrometry system. The invention provides an ion source for providing radiative heating to an ionization region. The ion source includes a nanospray ionization device for producing ions and a conduit adjacent to the ionization device for receiving ions from the ionization device. The conduit includes a conductive material for providing indirect radiative heating to the ionization region. Direct radiative heating may also be provided using a heater in the conduit. The ion source may be used separately or in conjunction with the mass spectrometry system. When used in conjunction with a mass spectrometry system a detector may also be employed down stream from the device. A method for desolvating an analyte using the device is also disclosed.
