    摘要: Method for detecting and quantifying an analyte in a liquid sample, using a test strip and magnetic particles as a detectable label and based on the detection of an amount of magnetic particles which become bound to a reading zone of a test strip as a result of performing the method, said amount being linked through a function to the analyte content of the sample, wherein the magnetic particles exhibit a nonlinear magnetization characteristic, which test strip is made of porous material and is arranged inside a case, cartridge or the like to form an assay device, wherein in its part surrounding the reading zone, the case is made of a material that is permeable to magnetic field, wherein for the reading of the reaction, the assay device is positioned in a measure cell of a magnetic reading device which detects the amount of magnetic particles in the reading zone by submitting it to at least one excitation magnetic field having about 90% of its power within one frequency band or a plurality of frequency bands, and the reading device measures the induced magnetic response field outside said frequency band or said plurality of frequency bands.

    摘要: Methods for detecting the presence or absence of, and for quantifying, one set of cells in a mixed cell population of at least two sets of cells especially Rh positive cells in a mixed population with Rh negative cells, as is found in a fetal maternal hemorrhage (FMH).The magnetic particles coated with anti-D antibodies are reacted with the Rh positive fetal cells in Rh negative maternal blood followed by a specific separation and quantifying technique.Gravitational forces or magnetic forces are used to move reacted magnetic particles to isolate, distinguish and quantify cells differentiated by antigenic composition.Rh positive cell volume is correlated to the volume of the original blood sample as an indication of the number of doses of RhIG needed to be administered to the mother to prevent subsequent Rh immunization.

    摘要: Bodily fluid is analyzed for the presence of drugs of a selected panel of drugs in a simultaneous assay in which sample of the fluid is incubated with additional amounts of all drugs of the panel, antibodies specific to each of the drugs of the panel, and microparticles, the microparticles being divided into subsets, one subset for each drug in the panel and each subset distinguishable from the others. The incubation is performed in a liquid medium in which competitive binding occurs, the drugs in the sample competing with those added to the assay medium for binding to the antibodies. In one procedure, the added drugs are pre-coupled to the microparticles while the antibodies are not, and the incubation is followed by further incubating the microparticles with labeled ligands that have affinity for the antibodies. In an alternative procedure, the added drugs are not coupled to the microparticles but are pre-labeled, while the antibodies are pre-coupled to the microparticles, and the assay proceeds without further incubation. In both alternatives, the microparticles are ultimately recovered from the assay medium and from any unbound species, and the recovered microparticles are analyzed by flow cytometry to obtain indications of the presence of the various drugs in the sample in an inverse manner by detection of the label, each drug differentiable from the others by the distinguishing features of the microparticles.

    摘要: A system for enumeration of cells in fluids by image cytometry is described for assessment of target populations such as leukocyte subsets in different bodily fluids or bacterial contamination in environmental samples, food products and bodily fluids. Briefly, fluorescently labeled target cells are linked to magnetic particles or beads. In one embodiment, a small, permanent magnet is inserted directly into the chamber containing the labeled cells. The magnets are coated with PDMS silicone rubber to provide a smooth and even surface which allows imaging on a single focal plane. The magnet is removed from the sample and illuminated with fluorescent light emitted by the target cells captured by a CCD camera. In another embodiment, a floater having a permanent magnet allows the target cells to line up along a single imaging plane within the sample solution. Image analysis can be performed with a novel algorithm to provide a count of the cells on the surface, reflecting the target cell concentration of the original sample.
