摘要: Provided herein are methods for increasing the yield of an extracellular product synthesized by an organism cultured in a continuous aerobic fermentation system. The extracellular product yield is increased through the use of an organism modified to decreased production of polyhydroxyalkanoate, to increase production of the extracellular product, and to include promoters that can be inducible in response to nutrient limitation conditions. The extracellular product yield is also increased by operating the continuous fermentation system under particular nutrient limitation conditions. Also provided are non-naturally occurring organisms that have been modified for use with the provided methods, and extracellular products made using the provided methods.

摘要: Provided is engineered microorganisms expressing acetoacetyl-CoA reductase variants and a method for improving the yield of PHA. Compared with the wild-type acetoacetyl-CoA reductase represented by SEQ ID NO. 31, the variant has one or more of the following mutations: (1) mutation of valine at position 141 to isoleucine or leucine; (2) mutation of methionine at position 12 to threonine, serine, alanine, leucine, lysine or isoleucine; (3) mutation of isoleucine at position 194 to valine, leucine or methionine; (4) mutation of glutamic acid at position 42 to lysine, glutamine, leucine, aspartic acid, proline, threonine, asparagine, or histidine; and (5) mutation of phenylalanine at position 55 to valine, alanine or isoleucine. The variants and their coding genes can promote the synthesis and accumulation of PHA by the strain and increase the yield of PHA.

摘要: A method for recovering polyhydroxyalkanoates from a biomass is disclosed. According to the method, polynucleotide chains are cleaved by addition of an endonuclease. A lysing agent is used to disrupt cell walls of the microorganism cells and release the intracellular polyhydroxyalkanoates from the cells. Proteins are also degraded by addition of a peptidase. The polyhydroxyalkanoates are then separated from cellular debris of the cells. According to the present disclosure, this method is carried out without the use of organic solvents in the cleaving, lysing, and degrading steps.

摘要: The disclosure provides for methods and a system for storing energy in the form of a biopolymer.
