Each fluid volume may contain a first primer pair to amplify a first allele of a target, a second primer pair to amplify a second allele of the target, a first reporter including a first photoluminophore and providing a primer of the first primer pair, and a second reporter including a second photoluminophore and providing a primer of the second primer pair. Each of the first and second reporters may include an oligomer having a quencher, and the oligomer may be configured to base-pair with the primer of the first primer pair and the primer of the second primer pair. The first and second alleles may be amplified using the first and second primer pairs. Photoluminescence may be detected. A level of each allele may be determined.

摘要: Various embodiments relate generally to the field of nucleic acid amplification and detection, in particular isothermal nucleic acid amplification and the detection of the amplicons using designated detection probes. Moreover, various embodiments also relate to methods for determining the presence or quantity of a target nucleic acid molecule in a sample using isothermal amplification. The detection probe is a single-stranded probe that recognises a probe binding site within target amplicons. The detection probe comprises at least one 3′end nucleotide mismatch and a quencher-fluorophore pair at the opposite ends of the probe. Following hybridization of the detection probe to the target amplicons, a DNA polymerase with 3′-5′ exonuclease activity can cleave the detection probe at the 3′ end nucleotide mismatch to release a 3′-terminal probe fragment comprising the quencher or fluorophore, thus generating signals.
