Abstract: A biomarker signal amplifier amplifies chemical analyte binding and includes: a surface strand disposed on an analysis substrate and including an exchange region; a particle strand hybridized to the surface strand in an absence of a chemical analyte that preferentially hybridizes to the exchange region as compared with the particle strand, and the particle strand is dissociated from the surface strand when the surface strand is in a presence of the chemical analyte; and a reporter particle attached to the particle strand and disposed proximate to the analysis substrate when the particle strand is hybridized to the surface strand in absence of the chemical analyte and that changes the electrical potential of the analysis substrate depending on whether the particle strand is hybridized to the surface strand.

In an exemplary embodiment, the target nucleic acid is a coronavirus target nucleic acid.

Abstract: Kits and methods are provided that utilize a mesophilic strand displacing polymerase selected from Bsu DNA polymerase (large fragment) and Klenow in Loop mediated amplification (LAMP) at temperatures in the range of 34° C.-52ºC. This contrasts with 60° C.-65° C. required for standard Bst polymerase dependent LAMP. The reduced temperature of the LAMP reaction enables the use of other proteins that are temperature sensitive in a one-step reaction. For example, a Cas protein such as Cas12a may be used with a target nucleic acid specific guide RNA and optionally a reporter oligonucleotide containing a quencher and a fluorophore or lateral flow reagents to determine the presence of pathogens in a sample.

Abstract: Systems, kits, and methods for detecting and quantifying proteomic activity using DNA-encoded probes are provided, where the proteomic activity may be enzymatic activity or ligand binding affinity. Such systems and methods encode quantitative proteomic activity information into DNA sequence populations and utilize DNA-linked substrates or ligands as activity probes. The systems, kits, and methods that are directed to detecting ligand affinity further include crosslinking steps to ensure the integrity of the DNA-linked ligands during purification and washing. Signal detection involves the chemical manipulation of a probe population downstream of sample exposure and application of purifying, selective pressure for desired products. Selection-induced changes in DNA abundance between the initial pool and the purified pool indicate sample activity.
