An inlet system is provided for direct injection of the liquid sample using a capillary tube. The method and apparatus allow for 20 to 2000-fold improvement of the lower detection limit of an analyte in a liquid sample compared to a conventional liquid chromatography/mass spectrometer apparatus.

    摘要: A mass spectrometer having a matrix-assisted laser desorption ionization (MALDI) source which operates at ambient pressure is disclosed. The apparatus and method are disclosed to analyze at least one sample which contains at least one analyte using matrix-assisted laser desorption ionization (MALDI), which apparatus comprises: The present invention relates to an apparatus and a method for ionizing at least one analyte in a sample for delivery to a mass analysis device, comprising: (a) an ionization enclosure including a passageway configured for delivery of ions to the mass analysis device; (b) means to maintain said ionization enclosure at an ambient pressure of greater than 100 mTorr; (c) a holder configured for maintaining a matrix containing said sample in the ionization enclosure at said ambient pressure; (d) a source of laser energy including means associated with the ionization enclosure for directing the laser energy onto said matrix maintained by the holder at the ambient pressure to desorb and ionize at least a portion of the analyte in the sample, and (e) means for directing at least a portion of the at least one ionized analyte into the passageway. The ambient pressure (AP-MALDI) source is compatible with various mass analyzers, particularly with mass spectrometers and solves many problems associated with conventional MALDI sources operating under vacuum. Atmospheric pressure MALDI is described. The analysis of organic molecules or fragments thereof, particularly biomolecules, e.g., biopolymers and organisms, is described.

    摘要: A MALDI mass spectrometer directs a laser shot onto a MALDI sample to generate a sample spectrum which is analyzed to determine if the sample spectrum meets a predetermined criteria. If so, subsequent laser shots are directed to predetermined locations on the MALDI sample. In essence, if the analysis of a previous laser shot indicates that a nullsweet spotnull of the MALDI sample has be located, subsequent laser shots may be directed to areas proximate to the previous shot thereby allowing the sweet spot to be thoroughly sampled. A method of operating a MALDI mass spectrometer is also disclosed.

    摘要: An automated pipetting apparatus and method for forming sample spots on a support include a pipette. A robotic assembly moves a sample container, such as a multiwell microtiter plate, and the support to the pipette for receiving and dispensing liquid samples. The pipette draws a predetermined volume of the liquid sample from the sample container into the axial passage of the pipette and forms a pocket of a gaseous material above and forms a barrier material below the volume of the liquid sample. The liquid sample is drawn into the pipette a distance sufficient to form a space between the liquid sample and the outlet of the pipette and to contain the liquid sample completely within the pipette. The volume of the barrier material, which is typically air or other fluid, separates the liquid sample a suitable distance from the outlet end of the pipette to prevent the liquid sample from flowing outward under gravity and being drawn along the outer surfaces of the pipette by the surface tension of the liquid, thereby preventing or inhibiting evaporation and preventing deposits from forming on the outer surface of the pipette. The pipette dispenses the liquid sample with sufficient force so that substantially the entire volume of the liquid sample is dispensed from the pipette directly onto the support. The support can be, for example, a sample plate of a MALDI mass spectrometer. The pipette deposits the liquid samples to form sample spots on the sample plate for analysis by the mass spectrometer.

The target position can be located within an adverse environment including at least one of a high magnetic field, a vacuum system, a high pressure system, and a hazardous zone.
