An improved system can be used to examine a centrifuged sample of anticoagulated whole blood for evidence of blood borne rare events such as: circulating cancer cells; malarial parasites; other hemato-parasites; bacteria; and the like; and can also be used in the measurement of hematocrit and hemoglobin, as well as white cell and platelet count values in the centrifuged blood sample. The system includes a transparent blood sample tube and an insert that is placed in the tube. The insert floats on the packed erythrocyte layer in the centrifuged blood sample, and expands all of the layers above the packed erythrocyte layer. The insert also forces any blood borne rare events to the periphery of the blood sample in the tube where such events can be detected through the tube wall. The bottom end of the tube is closed by a closure which includes an outer sheath and an inner plug that projects up into the tube bore to a degree necessary to elevate at least the upper portion of the packed erythrocyte layer into which the insert has settled sufficiently so as to be detectable above the cap, and preferably examinable microscopically after the blood sample has been centrifuged. The outer sheath of the tube closure includes an outer annular surface which is provided with sensible indicia. The indicia are operable to enable an instrument or a person examining the sample to rotate the tube, and to determine the circumferential location in the tube of any suspicious objects noted in the blood sample.

A target analyte which may be found in a substance, such as a biological or environmental substance, is assayed by inoculating a media with a sample of the substance. The target analyte is a unique nucleotide sequence of the RNA or DNA of a suspect organism, or any nucleotide target, or a combination of nucleotide sequences thereof, which organism may be found in the substance. The media contains selected target analyte amplifiers which will result in the amplification of any target analytes which are present in the substance. The media may also contain one or more labeled analyte-specific materials (LASMs) which can migrate through the media. The nature of the media is such that it will support target analyte-copying but will not allow the target analyte to migrate extensively within the media. After the substance to be assayed is added to the media and any target analytes are amplified, the LASMs will migrate by diffusion to resultant target analyte units or colonies in the media, thereby creating intensely labeled localized areas in the media which can be visually or mechanically detected. Each of the intensely labeled areas in the media will be surrounded by low intensity label halos. The number of high intensity labeled sites in the media will be proportional to the concentration of target analytes in the substance.

The scale can be presented in a normal image when a simple lens magnification, such as a magnifying glass, is used to view the tube and slide; and can also be presented in mirror image when compound lens magnification, such as a microscope, is used to view the tube and slide.

Constituent layers are harvested from a centrifuged multi-constituent material in an evacuated glass or clear plastic tube which contains a float. When possibly contaminated materials, such as blood, are being tested, the use of an evacuated tube allows the measurements to be made without the technician being exposed to the blood. The tubes are large enough to hold approximately one ml of blood, and are filled with an inert gas at low pressure. The floats are formed with a through bore into which cell bands to be harvested will settle during centrifugation. The cell bands are stabilized by a layer of a flowable material which settles onto the plasma layer during centrifugation and forms a pellicle thereon. The cell layers to be harvested are aspirated from the float bore by way of a hypodermic needle or cannula inserted into the tube and float bore.

A patient's blood sample is incubated with an antigen and tested for lymphocyte response, i.e. an activation of lymphocytes and/or a conversion of lymphocytes to lymphoblasts, which indicates prior exposure of the patient to the antigen. A positive response indicates the presence of prior exposure to prior diseases or clinical conditions such as parasitic diseases, tuberculosis, salmonellosis, gonorrhea, fungal infections, rickettsial infections, Lyme disease or allergens. Whole blood from the patient is incubated with the antigen of the disease or condition for which the patient is being tested. After a suitable time, a fluorescent dye or colorant which has an affinity for a discriminant characteristic of the activated lymphocytes or lymphoblasts, such as: intracellular calcium; surface activation antigens such as transferrin receptor; HLA-Dr; Leu-23; and the like. The incubated blood is then drawn into a transparent tube containing a float which concentrates the buffy coat constituent layers upon centrifugation of the blood sample. The concentrated lymphocyte layer is then examined for fluorescence or coloration which is indicative of the presence of the activated lymphocytes or lymphoblasts, and their concentration. The fluorescence or coloration can be qualified and/or quantified by a reader instrument.

A patient's health is diagnosed by centrifuging blood samples in a transparent tube, which tube contains one or more groups of particles such as lyposomes or plastic beads of different densities for each group. Each group of density-defined particles carries antigens or antibodies which are specific to a complement antigen or antibody which may be in the blood sample being tested, and which are indicative of the patient's health. A label-tagged antibody which is specific to all bound antibody/antigen couples is added to the blood sample so as to form labeled antibody+antigen-antibody complexes (AAAC) in the blood sample. Upon centrifugation, the complexed particles will settle out in different areas in the tube according to the respective density of the particles, and the degree of label emission of the particle layers can enable qualitative or quantitative analyses of the blood sample to be made. Unbound labeled antibodies will be washed away from the complexed layers by the washing action of the descending blood cells during the centrifugation step. Unbound labeled antibodies will thus not interfere with the analysis.

A whole blood sample is placed in a tube such as a capillary tube, with a plastic float. The float is axially elongated and has a specific gravity which causes it to float in the packed red blood cells when the sample is centrifuged in the tube. The hemoglobin concentration of the packed red blood cells is measured by measuring the depth that the float sinks into the red cell layer, and then the hemoglobin concentration of the blood is calculated. The hemoglobin can be measured in this manner because virtually the only red cell component which contributes density to the red cells is the hemoglobin. The hemoglobin concentration of the whole blood is calculated by multiplying the mean corpuscular hemoglobin concentration of the packed red blood cells by the packed cell volume % (hematocrit) of the whole blood. All of the necessary calculations may be performed by a preprogramed microprocessor.
