摘要: The present disclosure generally relates to systems and methods for partitioning species. In some embodiments, such systems can be used for determining one or more types of cancer. For example, certain aspects are generally directed to aqueous multi-phase partitioning systems that can be used, for example, for distinguishing between different types of cancers, diagnosing subjects with cancer, or the like. In some cases, such systems can be identified using solvent properties such as the solvent dipolarity/polarizability difference (Δπ*), the solvent hydrogen bond donor acidity difference (Δα), the solvent hydrogen bond acceptor basicity difference (Δβ), and the electrostatic property difference (c) between two phases of the partitioning system. These may be within certain ranges in accordance with various embodiments. Additionally, these ranges can be precisely tuned by controlling the compositions of the phases of the partitioning system to produce novel partitioning systems that are unexpectedly efficient at distinguishing between even relatively minor structural changes of proteins or other species, which accordingly can be used to distinguishing different types of cancers, or for other applications.

摘要: This disclosure provides IR700-molecule conjugates and methods of their use to remove (e.g., separate or isolate) a target from a sample in vivo or from a subject in vitro. It is shown herein that exposure of IR700 to near infrared (NIR) light removes a portion of IR700, changing it from a hydrophilic molecule, to one that is hydrophobic, resulting in aggregation of IR700 and anything bound to it. For example, the disclosed IR700-molecule conjugates and methods provide photo-controlled ways to control the pharmacokinetics of a drug in vivo, and can be used to remove undesired agents from environmental or food samples or to isolate target molecules in a laboratory.

摘要: A process of using a fish plasma component in a nutrient medium for cell culture includes obtaining a fish that is a progeny of domesticated broodstock that are reared under consistent and reproducible conditions. Blood is obtained from the fish, and plasma is separated from the blood. One or more specific components of the plasma are then extracted, and cells are cultured in a nutrient medium using the one or more extracted plasma components, and none of any remainder of the plasma. The plasma and/or the plasma components is/are tested for presence and/or level of endotoxin. Extracting the one or more specific components of the plasma, and/or culturing the cells is only performed if the testing indicates an endotoxin level below a predetermined threshold. The cells cultured using the extracted one or more plasma components are other than fish cells.

摘要: Separation of the cellular components of whole blood, or other biological fluid, from plasma or serum can be achieved for assay analysis. A device for facilitating separation can include, for example, a capillary tube that accurately draws target blood volume, a pad that chemically interacts with red-blood cells, such that the red blood cells become chemically and/or physically trapped within pad material, a mechanism for plasma recovery from the pad upon diffusion or active mixing, and a dropper tip that facilitates dispensing the mixture onto a test device. The treatment of the cellular components can be performed prior to contact with a buffer solution, so release of the cellular components into the buffer solution is reduced or prevented. Additional filtration can be provided to filter any remaining cellular components in the mixture.

摘要: A polypeptide is provided specifically binding to CD34 molecule and use thereof, the polypeptide is selected from at least one of polypeptide 17 and polypeptide 19, amino acid sequences of the polypeptide 17 and the polypeptide 19 are as shown in SEQ ID NO: 1 and SEQ ID NO: 2 in sequence listing. The polypeptide provided by the present application can specifically bind to CD34, and can be produced by artificial synthesis or genetic engineering method. Compared with antibody, the polypeptide provided by the present invention has characteristics such as low molecular weight, easy preparation and less immunological rejection; and it has little toxic side-effects, its binding to the CD34+ cell won't obviously kill the target cell and inhibit its proliferation. The polypeptide provided by the present invention can be used as markers for CD34 positive expression cells, and also can be used as substitute of immunofluorescence CD34 antibody.

摘要: The present invention relates to a microfluidic separation device, a separation method using the same and a kit for separating circulating rare cells from blood using the same, and more particularly, to a microfluidic-based separation technology for fixing target particles of a sample, which have a specific affinity for magnetic nanoparticles, to a device by use of a magnetic material, and for isolating the sample from which the target particles have been removed. The present invention may be effectively applied to remove leukocytes from a blood sample in order to isolate circulating rare cells (CRCs), particularly circulating tumor cells (CTCs).
