    Abstract: A valve having a body having a first section and a second section; an extended flange attached to the second section of the body or disposed as an integral part of the second section of the body; an elongate bore extending through the body and having a proximal end and a distal end; a longitudinally displaceable plunger disposed in and extending along the bore, the plunger having a proximal end and a distal end and having a first position displaced toward the distal end of the bore and a second position displaced toward the proximal end of the bore; a diaphragm seal attached to the proximal end of the plunger and sealing the bore at the proximal end thereof; a gland seal sealing the bore at a location intermediate the diaphragm seal and the distal end of the bore; the plunger extending through and being sealingly secured to the gland seal; a fluid transfer opening in the bore between the diaphragm seal and the gland seal; longitudinal displacement of the plunger moving the diaphragm seal to open the bore, the gland seal stretching to accommodate the displacement of and maintain a seal about the plunger, a fluid flow path being established between the open proximal end of the bore and the fluid transfer opening, wherein longitudinal displacement of the plunger towards its first position moves the diaphragm to open the bore. The valve further comprises an extended flange having a surface that is approximately coplanar with a surface of the second position when the plunger is displaced toward the proximal end of the bore, creating a zero dead leg position.

    Abstract: The invention concerns an installation for treatment of biological liquid by chromatography, extending in a longitudinal direction and comprising a supply valve (20b), a supply pump (30) downstream of the valve, instrument members downstream of the pump including distribution valves (81a-c, 82a-c, 83a-c) and devices (78a-c, 85a-c, 86a-c) for measuring physico-chemical parameters of the liquid, chromatography columns (99a-c) downstream of the instrument members and pipes connecting the valve, the pump, the instrument members and the columns so as to form at least one supply line for biological liquid to treat of a treatment circuit of said installation, the chromatography columns being disposed relative to each other in a direction of extension generally transverse to said generally longitudinal direction of extension of the installation.

    Abstract: A porous electrospun polymeric nanofiber liquid filtration medium, such as an electrospun mats, used for the removal of viral particles (e.g., parvovirus) and other particles in the 18 nm to 30 nm size range from fluid streams, having a mean flow bubble point measured with perfluorohexane above 100 psi. The electrospun medium includes nanofibers having an average fiber diameter of about 6 nm to about 13 nm, and the nanofiber liquid filtration medium has a mean pore size ranging from about 0.01 μm to about 0.03 μm, a porosity ranging from about 80% to about 95%, a thickness ranging from about 1 μm to about 100 μm, and a liquid permeability greater than about 10 LMH/psi. The high porosity of the electrospun mats enable much higher water fluxes, thus reducing the time required to complete virus filtration steps on a fluid stream.

    Abstract: A depth filtration device for the clarification of biological fluids including a composite depth filter media having a nonwoven first layer integral with a second layer containing a polyaeryionitrile (PAN) fibers, a filter aid, and a wet-strength resin. The depth filter media exhibits increased creased binding capacity for soluble impurities such as DNA and host eel! proteins from biological/cell culture feed-streams during secondary clarification and low-level impurity clearance of harvested cell culture fluids, such as those used for the manufacture of monoclonal antibodies. The depth filter media additionally exhibits significantly lower flushing requirements, resulting in lower levels of organic, inorganic and bioburden extractables released, high dirt holding capacities and good chemical and/or radiation resistance.

    Abstract: A process for the primary clarification of feeds, including chemically treated flocculated feeds, containing the target biomolecules of interest such as mAbs, mammalian cell cultures, or bacterial cell cultures, using a primary clarification depth filtration device without the use of a primary clarification centrifugation step or a primary clarification tangential flow microfiltration step. The primary clarification depth filtration device contains a porous depth filter having graded porous layers of varying pore ratings. The primary clarification depth filtration device filters fluid feeds, including chemically treated flocculated feeds containing flocculated cellular debris and colloidal particulates having a particle size distribution of approximately about 0.5 μm to 200 μm, at a flow rate of about 10 litres/m2/hr to about 100 litres/m2/hr. Kits and methods of using and making the same are also provided.
