Use of an identification sequence that results in a detectable phenotype increases the efficiency of screening for seed and plants in which transgene sequences not linked to a gene of interest have segregated from the sequence encoding a gene of interest.

Pesticidal proteins are provided that combine two or more modes of action into a single molecule. These pesticidal proteins therefor exhibit greater efficacy and/or durability of resistance, for the purposes of pest control, and can be utilized for pest control by provision in the diet of a pest organism, or by topical application to crop plants and/or pests. Methods and compositions for producing and using such proteins are also provided.

The present invention is directed to controlling pest infestation by inhibiting one or more biological functions in an invertebrate pest. The invention discloses methods and compositions for use in controlling pest infestation by feeding one or more different recombinant double stranded RNA molecules to the pest in order to achieve a reduction in pest infestation through suppression of gene expression. The invention is also directed to methods for making transgenic plants that express the double stranded RNA molecules, and to particular combinations of transgenic pesticidal agents for use in protecting plants from pest infestation.

The invention provides novel methods and compositions for introduction, transfer or delivery of one or more biomolecules into wounded recipient plant cell(s). Methods for production of a wounded recipient cell culture and the creation of one or more mutations, edits, transgenic insertions, or other genetic changes in the recipient cell(s) are also provided. Product cells produced by such methods, and resulting cells and regenerated plants, plant parts, and progeny plants are further provided. This invention provides a method to autoregulate expression of a transgene susceptible to sRNA silencing by concomitantly transcribing RNA from DNA of a transgene and RNA from DNA from at least one sRNA silencing pathway gene. An aspect of the invention provides use of a recombinant DNA construct that includes DNA of a transgene and DNA of an sRNA silencing regulator. Also disclosed are transgenic cells and organisms having in their genome a recombinant DNA construct that includes DNA of a transgene and DNA of an sRNA silencing regulator.

Recombinant DNA constructs, for use in plants and plant cells, have site-specific recombination sites that allow assessing phenotypes and modes of action by over expression or suppression of endogenous genes. In an aspect, a single DNA construct can be switched between over expression and suppression by the action of a recombinase such as the Cre recombinase on constructs having lox recombination sites. Other useful recombination systems include the Flp/frt system, the R/Rs system, the Dre/rox system, and the GIN/gix system.
