摘要: The invention relates to identification and characterization of recombinant DNA and polypeptides for specific Bt toxin receptors. In particular, the Bt toxin receptors of the invention include those derived from the Lepidopteran super family including the species Trichoplusiani ni, Pseudoplusia includens, Helicoverpa zea, and Spodoptera frugiperda. The receptors of the invention further include those derived from the Coleopteran super family and particularly from the species Diabrotica virgifera virgifera. The recombinant DNA and polypeptides so provided are useful in the identification and design of novel Bt toxin receptor ligands including novel or improved insecticidal toxins for use in a variety of agricultural applications. Materials and methods for identifying novel toxins are also disclosed herein. The invention also provides methods for selecting toxins to combine to control insect populations by manipulating Bt toxin receptor.

摘要: The present disclosure relates to a comprehensive model for expression of recombinant peptides by Pichia pastoris. The model uses an easily controllable variable called ‘critical nutrient ratio’ for obtaining a right balance between product synthesis and it's degradation during the fermentation process. The extra cellular concentration of precursor could be increased by about 10 folds and the degradation constants could be reduced by about 10-20 folds for intracellular and extracellular cases respectively by controlling critical nutrient ratio and addition of soya flour hydrolysate and EDTA.

摘要: The present invention relates to the purification of vitamin K-dependent blood coagulation factors, such as Factor IX (FIX). In particular, the invention provides a method for purifying Factor IX having a desired content of gamma-carboxyglutamic acid from a sample comprising a mixture of species of said Factor IX having different contents of gamma-carboxyglutamic acid, said method comprising the steps of: (a) loading said Factor IX sample onto an immunoaffinity chromatography material coupled to a binding moiety for gamma-carboxyglutamic acid; (b) eluting said Factor IX; and (c) selecting a fraction obtained from said elution wherein the polypeptides in the fraction have the desired content of gamma-carboxyglutamic acids; characterized in that the total concentration of Factor IX within said sample exceeds the binding ability of the immunoaffinity chromatography material.
