    摘要: A method for preparing treated naturally occurring or synthetic particles using a blood cell processor is provided. The method comprises adjusting the blood cell processor to allow a dispersion of such particles to come into contact with a treatment material in an amount and for a period of time sufficient to modify all or a portion of the surface of such particles. In a preferred embodiment, the method comprises washing collected red blood cells; separating a selected volume of said washed red blood cells; and treating one portion of said separated washed cells with a selected proteolytic enzyme, to prepare a red blood cell reagent.

    摘要: The present invention provides labeled nucleotide probes and a procedure for synthesis of the labeled nucleotide probe by a coupling or joinder of two components of the probe, namely a target recognition sequence moiety and a signal generating moiety, through the use of a bridging complement that holds the two moieties in such a position as to allow formation of a sugar-phosphate chemical linkage between the 3' end of one moiety and the 5' end of the other moiety.

    摘要: A new immunoassay system is provided for the detection of ligands or ligand binding partners in solution in a heterogeneous format. The invention relies upon the detection of back scattered light from an evanescent wave disturbed by the presence of a colloidal gold label brought to the interface by an immunological reaction. The evanescent wave existing at the interface in turn is the result of a totally internally reflected incident light wave. Placement of the detector at a back angle above the critical angle insures a superior signal-to-noise ratio. Apparatus and methods for scanning, detecting and manipulating light including a scattered total internal reflectance immunoassay system are provided.

    摘要: A device is described for preparing a standardized concentration of washed particles suspended in a fluid. The preferred method is directed to the preparation from whole blood specimens of a standardized concentration of washed red cells in isotonic solution using a device comprising a test tube formed at the bottom end with a nipple-like receptacle having a predetermined volumetric capacity. Whole blood is mixed with isotonic solution in the device which is then centrifuged causing the red cells to be first washed and then packed into the receptacle. The liquid contents of the device are then decanted without disturbing the packed red cells. Thereafter, the red cells are reconstituted to the desired concentration by adding isotonic solution to a predetermined level in the tube and mixing the cells into the isotonic solution.

    摘要: Methods for determining the presence of antigens or antibodies in an aqueous sample or presence of antigens on the surface of cells. A preferred embodiment employs fluorescent antigens which compete with the sample antigens for antibody binding sites. The antibodies are deposited on a support surface means in alternating patterns. The surface means and fluorescence detector are translocated with respect to each other and a signal generated by the detection of the repeating pattern of fluorescence. The signal is analyzed by means of a gated integrator responsive to a gate track control means also located on the surface means. Immunoassay methods having increased sensitivity are thereby obtained.
