摘要: A system and method is provided for remote session control of IP Multimedia Subsystem (IMS) sessions. A remote media flow may be established as part of an origination process wherein the controller UE requests that the media flow is established on a remote media UE as part of the origination request. A remote media flow may also be established as part of a termination process wherein the controller UE identified in the termination request identifies a remote media UE on which a remote media is to be established. Embodiments also allow for the session control to be transferred from a first device to a second device, while maintaining one or more media flows on the first device.

摘要: Some embodiments herein provide compositions and methods for expressing secreted and cell-surface-bound polypeptides in a single cell. In some embodiments, secreted and cell-surface polypeptide are produced from a single polynucleotide. The polynucleotide can comprise a sequence (or sequence encoding a polypeptide) that mediates separation of a membrane anchor from the polypeptide. In some embodiments, a desired ratio of secreted to surface-bound polypeptide is obtained by selecting a sequence that mediates a desired level of separation of the membrane anchor from the polypeptide.

摘要: The invention provides systems and methods for the generation of lymphocytes having a unique antigen specificity. In a preferred embodiment, the invention provides methods of virally infecting cells from bone marrow with one or more viral vectors that encode antigen-specific antibodies for the production of, for example B cells and T cells. In some embodiments, the viral vectors include an IRES or 2A element to promote separation of, for example, the α subunit and β subunit of a T cell receptor (TCR) or heavy and light chains of a B-cell antibody. The resulting lymphocytes, express the particular antibody that was introduced in the case of B cells and TCR in the case of T cells. The lymphocytes generated can be used for a variety of therapeutic purposes including the treatment of various cancers and the generation of a desired immune response to viruses and other pathogens. The resulting cells develop normally and respond to antigen both in vitro and in vivo. We also show that it is possible to modify the function of lymphocytes by using stem cells from different genetic backgrounds. Thus our system constitutes a powerful tool to generate desired lymphocyte populations both for research and therapy. Future applications of this technology may include treatments for infectious diseases, such as HIV/AIDS, cancer therapy, allergy, and autoimmune disease.

摘要: Provided are methods for generating immune cells of the desired type and specificity in a host. The methods may be used to treat a disease or disorder, such as a tumor in a patient. Target cells, preferably hematopoietic stem cells such as primary bone marrow cells are transfected with a polynucleotide encoding a T cell receptor with the desired specificity. The transfected cells are then transferred to the host where they develop into mature, functional immune cells. The source of the T cell receptor can determine the stem cell's fate. Thus transfecting stem cells with TCRs from cytotoxic cells will lead to the generation of cytotoxic T cells in the host, while TCRs from helper cells will produce helper cells. Both arms of T cell immunity can be generated simultaneously in a host. Additionally, the immune response to the desired antigen can be further stimulated by immunizing the host with the antigen.
