Compositions are disclosed, comprising dendrimers to which a first polypeptide is controllably coupled. Such polypeptide-dendrimer compositions are effective for controllably coupling a second polypeptide to the dendrimer. The first and second polypeptides have separate and distinct defined biological activities, for example, two antibodies with first and second binding specificities or an antibody and an enzymatic label. Such compositions are useful as indicators in specific binding assays, e.g., immunoassays. Methods for sequentially coupling two different polypeptides to a dendrimer to form compositions of the invention also are disclosed.

Methods, compositions and articles of manufacture are provided for conducting specific binding assays to determine the concentration or presence of at least one analyte in a sample. At least two dendrimer-reagent preparations with different analyte specificities may be immobilized on a solid phase. Alternatively, at least one dendrimer-reagent preparation having multiple specificities may be immobilized on a solid phase. Immobilization is facilitated by coupling specific binding assay reagents such as polypeptide receptors or analytes with water soluble polymers. The antibody reagent is a conjugate of a small molecule attached by a spacer group to an antibody for the analyte. The method comprises controlling, in the preparation of the conjugate, reaction parameters comprising the hydrophobicity or hydrophilicity of the spacer group, the length of the spacer group, the number of molecules of the small molecule attached to the antibody and the point of attachment of the small molecule to the antibody to obtain an optimum assay sensitivity and/or dynamic range. In some embodiments the method comprises preparing two or more conjugates by selecting a set of parameters for each conjugate wherein the set of parameters is different for each conjugate, conducting an assay for the analyte employing each conjugate and selecting for use in the assay the conjugate that provides the optimum assay sensitivity and/or dynamic range.

The present application relates to a method of making liposomes having membrane proteins incorporated therein, the method comprising: providing the membrane protein in solution; providing a solution of preformed liposomes; and incubating the mixture. Prior to the step of providing a solution of preformed liposomes, the liposomes are formed by combining a mixture of phospholipids with a solution of at least one type of unsaturated fatty acid. The methods of the present invention further relate to the method of making a reagent comprising tissue factor reconstituted into preformed liposomes. The method of the present invention for making a tissue factor reagent comprises: providing tissue factor in solution; providing a solution of preformed liposomes comprising a mixture of phospholipids and at least one type of unsaturated fatty acid; and incubating the mixture.

The storage device executes the command and sends the result to the computing device. As a result, the data is not sent to the computing device for manipulation.
