摘要: Methods of treating a subject in need thereof are provided which include: administering a recombinant platelet derived growth factor D (PDGF D) composition to a mesenchymal stem cell of the subject and/or a progenitor derived therefrom, in vivo, or ex vivo, producing a treated mesenchymal stem cell of the subject and/or a progenitor derived therefrom, thereby stimulating the mesenchymal stem cell (MSC) and/or a progenitor derived therefrom. Cells expressing a recombinant PDGF D composition of the present are administered to the subject for in vivo delivery of the recombinant PDGF D composition according to aspects of the present disclosure. Methods and compositions are provided including recombinant PDGF D hemidimer (HD) including a full-length PDGF D polypeptide and a C-terminal growth factor domain of PDGF D, which lacks a CUB domain, promoting regulation of bone marrow MSC differentiation into osteogenic lineage cells.

摘要: The present disclosure relates to targeted degradation platform technology. For example, the present disclosure relates to bispecific binding agents for degrading endogenous proteins, whether membrane-associated or soluble, using the lysosome pathway. The disclosure also provides methods useful for producing such agents, nucleic acids encoding same, host cells genetically modified with the nucleic acids, as well as methods for modulating an activity of a cell and/or for the treatment of various disorders.

摘要: Cell penetrating peptides that target many cells types including cells of the retina and cornea with high efficiency are provided herein. These peptides can be used to deliver cargo molecules across a plasma membrane, without the need for chemical conjugation. Compositions and viral vectors comprising these cell-penetrating peptides are also provided. Methods of using the peptides, compositions and viruses to deliver various agents to target cells and tissues are also provided.

摘要: The invention relates to apH mediated cell delivery vehicle comprising: a cargo or a cargo-binding molecule for binding to a cargo; a protein transduction domain; and a glycosaminoglycan (GAG) binding element, which is capable of binding to GAG on the surface of the cell, wherein the GAG binding element is a peptide which is modified to comprise one or more histidine residues which are capable of being protonated in an acidic environment. The invention further relates to the use of such a delivery vehicle including methods of treatment, such as treatment for cancer.

摘要: Methods are disclosed herein for increasing bone mass and strength or bone fracture healing in a subject. The methods include administering to the subject a therapeutically effective amount of multipotent stem cells, wherein each multipotent stem cell is transformed with a recombinant nucleic acid molecule comprising a heterologous promoter operably linked to a nucleic acid encoding platelet derived growth factor (PDGF) B, and wherein the multipotent stem cells express a sufficient amount of PDGFB to increase bone mass and strength or bone fracture healing. A lentiviral vector also is disclosed that includes a phosphoglycerate kinase-1 (PGK) promoter operably linked to a nucleic acid encoding PDGFB.

摘要: A method of increasing blood-brain barrier permeability of selected brain tissue in a subject in need thereof is carried out by: (a) parenterally administering to the subject stem cells that migrate to the brain tissue, the stem cells containing a recombinant nucleic acid, the recombinant nucleic acid comprising a nucleic acid encoding a barrier-opening protein or peptide operably associated with a heat-inducible promoter; and then (b) selectively heating the selected brain tissue sufficient to induce the expression of the barrier-opening protein or peptide in an amount effective to increase the permeability of the blood-brain barrier in the selected brain tissue. Nucleic acids, vectors, stem cells and compositions useful for carrying out such methods are also described.

摘要: A composition including an elastin-like polypeptide (ELP) coupled to a therapeutic agent is provided. The ELP comprises at least about 5 repeats of the amino acid sequence VPGXG. Further provided is a method of using the composition for therapeutic agent delivery during pregnancy to reduce the amount of the therapeutic agent crossing a placenta in a pregnant subject. The method includes administering to the pregnant subject an effective amount of the composition comprising the ELP coupled to the therapeutic agent.
