Abstract: A two-step chromatography purification scheme is described which selectively captures and isolates the genome-containing rAAV vector particles from the clarified, concentrated supernatant of a rAAV production cell culture. The process utilizes an affinity capture method performed at a high salt concentration followed by an anion exchange resin method performed at high pH to provide rAAV vector particles which are substantially free of rAAV intermediates.

Abstract: Chromatography devices contain chromatography media and methods of making and methods of using chromatography devices. Chromatography devices enable a more efficient, productive and/or environmentally friendly chromatographic operation due to one or more of the following advantages over conventional chromatographic operations: elimination of a device packing step by the user; elimination of clean-in-place (CIP) steps; elimination of clean-in-place (CIP) steps utilizing sodium hydroxide solution; elimination of any validation steps by the user; and use of a chromatography device comprising biodegradable material. The chromatography media includes porous inorganic particles having a functionalized surface and having a median pore size of at least about 300 Angstroms (A), or at least about 300 A up to about 3000 A. The inorganic particles may have a BET surface area of at least about 20 m2/g, or at least about 25 m2/g, or about 30 m2/g, up to about 2000 m2/g.

Abstract: A method for modifying a polymer carrier material for use as a stationary phase in an analytical or preparative separating method, the method comprising the steps of: providing a polymer carrier material, which is at least partly formed of aromatic hydrocarbon compounds comprising at least two vinyl or allyl substituents; producing hydroxy groups on/in the polymer carrier material by a method comprising an oxidative treatment of the polymer carrier material and a subsequent reductive or hydrolytic treatment of the reaction product; reacting the product from the previous step with a polyfunctional compound. The invention also relates to a polymer carrier material for use as a stationary phase in an analytical or preparative separating method, in particular a chromatography method, produced according to a method according to the invention.

Abstract: Open tubular capillary columns for liquid and ion chromatography, based upon an ionically impermeable polyolefin capillary having a bore with a sulfonate-group- or amine-group-functionalized internal surface. The capillary columns may include a coating of ion exchanging nanoparticles electrostatically bound to the functionalized internal surface. The capillary columns may be made by exposing the interior surface to a sulfonating reagent comprising chlorosulfonic acid (CISO3H), preferably from 85 wt % to 95 wt % chlorosulfonic acid at a process temperature of 20 to 25° C. The interior surface may be subsequently exposed to an asymmetrical diamine to form a sulfonic mid-linkage to the diamine, i.e., to form a sulfonamide-linked, amine-group-functionalized internal surface. The coating may be provided by subsequently exposing the interior surface to an aqueous suspension of ion exchanging nanoparticles to electrostatically bond the ion exchanging nanoparticles to the functionalized internal surface.
