    摘要: A novel nucleotide derivative, in case of existing as a member of a single-stranded sequence, undergoing a change in the fluorescent signal intensity depending on the corresponding base type in the partner strand with which the single-stranded sequence is hybridized, and which is a thymine/uracil derivative (1) emitting light most intensely when a confronting base in the partner strand with which the single-stranded nucleotide sequence is hybridized is adenine; a cytosine derivative (2) emitting light most intensely when the confronting base is guanine; an adenine derivative (3) emitting light most intensely when the confronting base is cytosine; a guanine derivative (4) emitting light most intensely when the confronting base is cytosine or thymine/uracil; and an adenine derivative (5) emitting light most intensely when the confronting base is thymine/uracil.

    摘要: The present teaching provides a fluorescent oligonucleotide probe having a high degree of design flexibility and wide applicability, as well as the use thereof. This is an oligonucleotide probe capable of forming a stem and loop, comprising at least one fluorophore located between adjacent nucleotides in the stem and is linked to a unit represented by Formula (1) and at least one quencher located at a site capable of pairing up with the at least one fluorophore located between the adjacent nucleotides in the stem and is linked to a unit represented by Formula (2).     摘要: An object of the disclosure of the present specification is to provide a method for detection of a target nucleic acid which allows construction of an effective detection system of a target nucleic acid. For this purpose, in the disclosure of the present specification, a first primer comprising an identification sequence complementary to a target sequence in a target nucleic acid and a tag addition sequence, and a second primer having a label are prepared. The first primer and the second primer are used for the target nucleic acid in a sample to amplify a chimeric DNA having a tag sequence and the label. The chimeric DNA is hybridized with a detection probe on a solid phase to obtain signal intensity information based on the label, and the target nucleic acid is detected based on the signal intensity information.

    摘要: In a state where liquids are contained in a plurality of liquid container portions which are formed in a first layer and a second layer of a cartridge and which have predetermined volumes determined depending on liquids to be contained, and where the reaction vessel is connected to any one of communicating ports, when pressure is applied to act upon one of the liquid container portions which is communicated with the communicating port connected to the reaction vessel, the atmosphere is supplied to the one liquid container portion through a atmosphere flowing passage communicating with the outside, and the liquid contained in the one liquid container portion is supplied to the reaction vessel. The cartridge is rotated to connect another communicating port to the reaction vessel such that plural liquids are eventually supplied to the reaction vessel. On that occasion, since the atmosphere supplied through the atmosphere flowing passage is further supplied to the reaction vessel through the communicating port connected to the reaction vessel and the one liquid container portion, the plural liquids contained in the reaction vessel are stirred by the inflow of the atmosphere.

    摘要: A method of detecting or analyzing a target sequence in a genomic DNA by using a capture probe immobilized on a solid carrier includes: bringing the target nucleic acid into contact with a first query probe that has a sequence complementary to a portion of the target sequence or to a sequence adjacent to the portion and a second query probe that has a sequence complementary to another portion of the target sequence or to a sequence adjacent to the another portion and that has a recognition sequence complementary to a portion of the capture probe; acquiring a ligated molecule by ligating the first query probe and the second query probe that are hybridized to the target nucleic acid; bringing the ligated molecule into contact with the capture probe on the solid carrier and then capturing the ligated molecule on the solid carrier by hybridizing the capture probe with the recognition sequence in the ligated molecule; and detecting the captured ligated molecule.
