    摘要: A method of amplifying one or more target nucleic acid sequence(s) in a PCR buffer solution comprising one or more target nucleic acid sequence(s), a nucleic acid polymerase, deoxynucleotide or nucleotide triphosphates, at least a first primer and a second primer is provided. The PCR buffer solution is contained in an elongated vessel. The solution within the vessel is locally heated in a spatio-temporally varying manner. Fluid flows circulating within the vessel are established. At least a fraction of the PCR buffer solution cyclically travels between regions having different temperatures, giving rise to a temperature cycling.

    摘要: The present invention pertains to a method and a device for the determination of thermo-optical properties, particularly the size or size distribution, of fluorescently labeled biomolecules or biomolecule complexes, particularly nucleic acids, in a reaction solution. The method comprises the steps of: (i) providing a reaction solution with fluorescently labeled biomolecules or biomolecule complexes; (ii) irradiating a laser light beam into the solution to obtain a spatial temperature distribution in the solution around the irradiated laser light beam; (iii) exciting fluorescently said fluorescently labeled biomolecules and detecting the fluorescence at two or more defined regions representing different mean temperatures in said spatial temperature distribution, wherein said detection of fluorescence is performed at least once at a predetermined time after the start of the laser irradiation; and (iv) determining the thermo-optical properties, particularly the size or size distribution, of the fluorescently labeled biomolecules or biomolecule complexes from the detected fluorescence intensity or fluorescence intensity distribution.

    摘要: A polymerase chain reaction apparatus includes: a substrate; a high-temperature sidewall erected on the substrate; a low-temperature sidewall erected on the substrate and facing the high-temperature sidewall; and a reaction chamber consisting of the substrate, the high-temperature sidewall, and the low-temperature sidewall, wherein a sample contained in the reaction chamber is repetitively thermal-circulated between the high-temperature sidewall and the low-temperature sidewall using Marangoni convection generated by a surface tension gradient resulting from a temperature difference in an interface between the sample and air. The PCR amplification can be automatically accomplished by surface tension flow generated by Marangoni convection resulting from a temperature difference in an interface between the sample and air when a temperature difference between the sidewalls of the chamber is maintained constant. As a result, it is possible to reduce power consumption, simplify the configuration of a temperature control circuit, and reduce the time for a cycle of amplification.
