摘要: In Multi-Policy Decision-Making (MPDM), many computationally-expensive forward simulations are performed in order to predict the performance of a set of candidate policies. In risk-aware formulations of MPDM, only the worst outcomes affect the decision making process, and efficiently finding these influential outcomes becomes the core challenge. Recently, stochastic gradient optimization algorithms, using a heuristic function, were shown to be significantly superior to random sampling. In this disclosure, it was shown that accurate gradients can be computed—even through a complex forward simulation—using approaches similar to those in dep networks. The proposed approach finds influential outcomes more reliably, and is faster than earlier methods, allowing one to evaluate more policies while simultaneously eliminating the need to design an easily-differentiable heuristic function.

摘要: A two-part topical composition includes a first composition including a S-nitrosothiol nitric oxide donor and a carrier, and a second composition including zinc oxide. The second composition is to be mixed with the first composition. The zinc oxide reacts with the S-nitrosothiol nitric oxide donor at a temperature ranging from about 20° C. to about 40° C. to enhance a rate of release of nitric oxide from a mixture of the first composition and the second composition.

摘要: Methods and systems perform magnetic resonance fingerprinting (MRF) by obtaining magnetic resonance data over a main field-of-view (FOV) and resulting from providing a magnetic resonance fingerprinting pulse sequence to a sample. The pulse sequence includes gradient waveforms and radio frequency (RF) pulses that have pulse sequence parameters specifically tailored for scanning, not the entire main FOV but rather a reduced portion of that main FOV. The methods and systems further include comparing the magnetic resonance data from the sample to a fingerprint dictionary of signal profiles that specifically correspond to the reduced portion of the main FOV and generating tissue property maps that correspond only to that reduced portion.

摘要: A method of estimating lung motion includes collecting multiple ultrasound image data captured at one or more locations of a sample region of tissue. The method further includes comparing the multiple ultrasound image data and determining temporal correlation coefficients between each of the multiple ultrasound image data. The method still further includes displaying an image of the sample region of the tissue with the temporal correlation coefficients identified, thereby indicating lung motion. In further methods, the determined temporal correlation coefficients are used to determine an amount of decorrelation, which can be used to determine strain of the tissue over the sample region and to calculate lung displacements and lung shape changes representing ventilation.

摘要: Methods comprising an integrated, multiscale artificial intelligence-based system that reconstructs drug-specific pharmacogenomic networks and their constituent functional sub-networks are described. The system uses features of the functional topology of the three-dimensional architecture of drug-modulated spatial contacts in chromatin space. Discovery of a drug pharmacogenomic network is made through the selection of candidate SNPs by imputation, determination of the predicted causality of the SNPs using machine learning and deep learning, use of the causal SNPs to probe the spatial genome as determined by chromosome conformation capture analysis, combining targeted genes controlled by the same cell and tissue-specific enhancers, and reconstruction of the pharmacogenomic network using diverse data sources and metrics based on the results of genome-wide association studies. Knowledge-based segmentation methods are used to deconstruct the pharmacogenomic network into its constituent efficacy and adverse event sub-networks for applications in clinical decision support, drug re-purposing, and in silico drug discovery.

摘要: The present disclosure relates to fluorescent biosensors and methods of use thereof. In particular, provided herein is a genetically encoded fluorescent indicator (GEFI) comprising a circular-permuted fluorescent protein (cpFP) and an inhibitory molecule bound to the cpFP. In the basal state, the inhibitory molecule prevents fluorescence from the circular-permuted fluorescent protein. Upon conformational change and/or cleavage of the bond between the inhibitory molecule and the cpFP, the cpFP is disinhibited, thereby permitting fluorescence from the cpFP. The biosensors described herein may be used in methods for detecting a variety of cell-signaling events.
