    摘要: The inventors have discovered that an ATPase-deficient dominant-negative mutant NS3 protein of hepatitis C virus inhibits activity of the wild-type NS3 protein and inhibits replication of hepatitis C virus (HCV). The solved crystal structure of a multi-enzyme NS3 complex on a DNA substrate is also provided. The inventors have tested a peptide matching the sequence of a portion of NS3 that interacts with another NS3 molecule for inhibiting HCV replication. The peptide inhibits HCV replication. Accordingly, the invention provides a method of inhibiting HCV replication in cells infected with HCV involving transforming the cells with a vector expressing a dominant-negative mutant NS3 gene. The invention also provides a method of inhibiting HCV replication in cells infected with HCV involving administering to the cells a dominant-negative mutant NS3 protein. The invention also provides peptides and agents that inhibit HCV replication and methods of identifying agents that inhibit HCV replication.

    摘要: Gene expression profiling reveals four distinct subgroups of multiple myeloma that have significant correlation with various clinical characteristics. Diagnosis for multiple myeloma (and possibly monoclonal gammopathy of undetermined significance) based on differential expression of 14 genes, as well as prognosis for the four subgroups of multiple myeloma based on the expression of 24 genes are established. A 15-gene model that classifies myeloma into 7 groups is also reported. Gene expression profiling also allows placing multiple myeloma into a developmental schema parallel to that of normal plasma cell differentiation. Development of a gene expression- or developmental stage-based classification system for multiple myeloma would lead to rational design of more accurate and sensitive diagnostics, prognostics and tumor-specific therapies for multiple myeloma.

    摘要: The invention involves the discovery that Chlamydia sp. strains can be cured of their plasmids by treatment with novobiocin, and that plasmid-deficient strains are defective in infecting cells under standard conditions, but can infect cells if centrifuged onto the host cells. But it is found that plasmid-deficient strains with wild-type infection efficiency under standard conditions can be isolated as mutants from parent plasmid-deficient strains with low infectivity by selecting for infection under standard conditions. Both the less infective and the highly infective plasmid-deficient strains were able to infect mice with little or no pathological symptoms, and both reduced the pathology in mice later challenged with the parental wild-type disease-causing Chlamydia strain. Thus, plasmid-deficient Chlamydia are effective vaccine strains. The invention provides a process for isolating a plasmid-deficient strain of Chlamydia sp., a process for developing a plasmid-deficient strain of Chlamydia sp. for use as a vaccine, a process for developing a highly infective plasmid-deficient strain of Chlamydia sp., isolated Chlamydia sp. strains, a method of expressing a recombinant nucleic acid in Chlamydia sp., and a method of immunizing a mammal against a Chlamydia infection.

The treatment amount can be effective to reduce phosphorus solubility in the manure; reduce phosphorus runoff and/or phosphorus leaching from fields fertilized with manure; inhibit ammonia volatilization from the manure; flocculate solids in the manure; reduce pathogens in the manure; increase the nitrogen content in the manure; and/or reduce acid rain and PM-10s associated with the manure.
