Novel bipyridyl-osmium complex conjugates and their use in electrochemical assays are described. The redox reversible-osmium complexes can be prepared to exhibit unique reversible redox potentials and can thus be used in combination with other electroactive redox reversible species having redox potentials differing by at least 50 millivolts in electrochemical assays designed for use of multiple electroactive species in the same cell and in the same sample without interference between the two or more redox coupled conjugate systems.

These methods comprise contacting a sample with a reagent labeled with an electrochemiluminescent chemical moiety and capable of combining with the analyte of interest, exposing the resulting sample to electrochemical energy and detecting electromagnetic radiation emitted by the electrochemiluminescent chemical moiety.

A method and device for detection and quantification of biologically significant analytes in a liquid sample is described. The method includes contacting a volume of a liquid sample with predetermined amounts of at least a first and second redox reversible species having redox potentials differing by at least 50 millivolts. At least one of the redox reversible species comprises a liquid sample diffusible conjugate of a ligand analog of an analyte in the liquid sample and a redox reversible label. A predetermined amount of at least one specific binding partner for each analyte to be measured is combined with the sample and current flow is measured at first and second anodic and cathodic potentials and correlated with current flows for known concentrations of the respective diffusible redox reversible species. Diagnostic devices and kits, including such devices and the specified specific binding partner(s) and redox reversible species are also described.

Electrochemiluminescent assay methods for determining an analyte of interest, which methods include forming one or more compositions which include, in the aggregate, (a) a sample to be tested for the analyte of interest, (b) a component capable of specifically binding with the analyte, (c) a label reagent which, when oxidized, is capable of electrochemiluminescence, (d) an amine which, when oxidized, forms a reducing agent, and (e) an electrolyte solution; subjecting such a composition containing the sample to conditions sufficient for specific binding to occur between the analyte of interest, if present, and one or more of the other components in the composition; thereafter, a composition containing the label reagent and amine, and reflecting the outcome of applying the aforementioned binding conditions, is exposed to conditions, such that both the label reagent and the amine are oxidized, the amine forms a reducing agent which interacts with the label reagent, electrochemiluminescence occurs, and the luminescence emitted from the label reagent is used to determine the analyte of interest.

A chemical moiety is disclosed which comprises a chemical biochemical, or biological substance attached to one or more electrochemiluminescent organometallic compounds. In a preferred embodiment of the invention the substance is attached to one or more ruthenium-containing or osmiumcontaining luminescent organometallic compounds. Methods are disclosed for detecting very small amounts of the chemical moiety using chemiluminescent, electrochemiluminescent, and photoluminescent means. Compounds are disclosed which are useful for labelling substances of interest with ruthenium-containing and osmium-containing labels or other electrochemiluminescent labels. These labelled substances are useful in methods provided for detecting and quantifying analytes of interest in binding assays and competitive binding assays. The labelled substances are of particular use in homogeneous binding assays. These methods form the bases for systems designed to enable the rapid, efficient, and sensitive determination of a broad array of chemical, biochemical, and biological materials of interest.

A complex of a tetradentate tetraanionic ligand L of the formula: ##STR1## and a metal, preferably a transition metal, is described. The chelate and ring Z-CH-M-Z functional groups Z, such as N or O are resistant to oxidation and the ring contains 4 to 7 preferably 5 to 6 chain atoms. An osmium complex containing two pyridine ligands is an oxidation catalyst. Further activation by anodic oxidation and heating to a temperature exceeding 30.degree. C. greatly increases catalytic activity.

A process for producing linear aldehydes or alcohols from olefins using a transition metal hydroformylation catalyst wherein the catalyst comprises an anionic transition metal catalyst wherein the transition metal is Fe, Ru or Os and the catalysts generally have a charge of at least -2. The formula of the anionic catalyst is generally defined as M.sup.+n [H.sub.y A.sub.x L.sub.z ].sup.-n wherein A represents Fe, Ru or Os, M is a cationic species, n is an integer greater than or equal to 2, x is an integer greater than or equal to 1, y is an integer greater than or equal to 0 and z is an integer generally corresponding to the number of available coordination bonding sites in the ruthenium complex. Monoanionic osmium catalysts are also included within the present invention. These hydroformylation catalysts exhibit greater selectivity toward straight chained aldehydes and alcohols than anionic catalysts having a charge of -1 or a neutral catalyst. The monoanionic osmium catalyst also exhibits an increased selectivity toward the straight chain product.The novel hydroformylation catalyst of the present invention is also disclosed.
