    摘要: Qualitative and quantitative electrochemiluminescent assays for analytes of interest present in multicomponent liquids are provided. These methods comprise contacting a sample with a reagent labeled with an electrochemiluminescent chemical moiety and capable of combining with the analyte of interest, exposing the resulting sample to electrochemical energy and detecting electromagnetic radiation emitted by the electrochemiluminescent chemical moiety. Further provided are methods for detecting and identifying the presence of a multiplicity of analytes in a liquid food or food homogenate. These methods comprise immersing a diagnostic reagent holder, provided with a multiplicity of reagents, into the food or food homogenate, removing the diagnostic reagent holder from the liquid food or food homogenate, and detecting and identifying the presence of a multiplicity of analytes of interest bound to the diagnostic reagent holder, thereby detecting and identifying the presence of a multiplicity of analytes of interest in the food or food homogenate. The invention further provides an enzyme immunoassay for coliform bacteria. This assay comprises inoculating a sample into a suitable medium for coliform reproduction, immobilizing coliforms present in the medium to a suitable surface, treating the surface with an antibody directed to the immobilized coliforms and detecting the presence of the immobilized coliforms immobilized to a suitable surface.

N-azoles, which are especially useful as secondary enhancers are imidazole, 1-methylimidazole, 1,2,3-triazole and 1,2,4-triazole. The invention also describes the use in diagnostic assays of chemiluminescent substrates containing said N-azoles, as secondary enhancers.

    摘要: A method for determining the concentration of heparin in a fluid sample comprising: (a) providing a fluid sample containing heparin; (b) adding to the fluid sample a solution of a dye to form a mixture of sample and dye, wherein the dye interacts with the heparin in the sample so that the absorption spectrum of the mixture of sample and dye in the visible range varies as a result of the interaction in a manner quantitatively dependent on the heparin concentration; (c) determining the absorption spectrum in the visible range of the mixture of sample and dye; and (d) calculating a spectral parameter representative of both the reduction in the absorption of the free dye in solution and the increase in the absorption of the dye-heparin complex, the value of the spectral parameter being indicative of the concentration of heparin present in the fluid sample, in order to determine the concentration of heparin present in the fluid sample. The relationship between the value of the spectral parameter and the concentration of the heparin present in the fluid sample has been previously determined by determining the absorption spectra, in the visible range, of a composition comprising the fluid and the dye, in the absence of heparin and in the presence of a plurality of concentrations of heparin, and calculating the relationship between the value of the spectral parameter and the concentration of heparin. A device for performing this method is also provided.

    摘要: The present invention relates to a kit for the labelling of biomolecules bearing reactive amino or hydroxyl groups. The kit consists of a Reagent A and a Reagent B, individually packaged, comprising: a mixture of a carboxylated labelling compound and a tertiary amine (Reagent A); and a coupling reagent (Reagent B). Upon contacting Reagent A with Reagent B, the carboxylated labelling compound is activated in-situ by the coupling reagent (a guanidinium, or uranium salt) in the presence of the tertiary amine. The active form of the carboxylated labelling compound is reacted with a biomolecules bearing reactive amino or hydroxyl groups, with formation of a stable covalent bond between the carboxylated labelling compound and the biomolecule.
