摘要: The present disclosure provides an N protein epitope mutation marker for preparing an epitope deletion-marked vaccine strain of type II porcine reproductive and respiratory syndrome virus (PRRSV) and use thereof, belonging to the technical field of biological products. In the mutation marker, one or more amino acids are mutated based on an epitope sequence at positions 92 to 103 of a C-terminal of an N protein of the type II PRRSV; and the epitope mutation marker has an amino acid sequence shown in SEQ ID NO: 1, where X1 is selected from the group consisting of T, P, and A; and X2 is selected from the group consisting of V and A.

摘要: The present disclosure belongs to the technical field of African swine fever (ASF) treatment, in particular to use of a compound aprepitant in prevention or treatment of the ASF. In the present disclosure, it is found that the compound aprepitant can significantly inhibit replication of an African swine fever virus (ASFV); in addition, it is found that the aprepitant inhibits transcription and protein expression of D1133L, and reduces transcription and protein expression levels of p30 and p72, preventing virus invasion on host cells. Therefore, the compound aprepitant can be used to inhibit early and late infection of the ASFV. Accordingly, the compound aprepitant can be used to prevent or treat the ASF.

摘要: The present disclosure relates to a cocktail vaccine of a recombinant African swine fever virus (ASFV) antigen and use thereof, and belongs to the field of biotechnology pharmacy. The cocktail vaccine is prepared by combination of recombinant proteins p30-modified p54, p72 epitope-N-segment amino acid sequence of pE248R, and N-terminal amino acid sequence of CD2v-C-terminal amino acid sequence of pEP153R with an adjuvant. The present disclosure provides a method for expressing and purifying the three recombinant proteins, as well as a ratio of each recombinant protein and the adjuvant for preparing the cocktail vaccine.

摘要: The present invention relates to a method for preparing a whole bovine-derived broadly neutralizing antibody against serotype O foot-and-mouth disease virus, and belongs to the technical field of antibody preparation. The method includes the following steps: 1) conducting the immunization on the cattle; 2) screening for the serotype O foot-and-mouth disease virus antigen-specific single B cells; 3) amplifying variable region genes of the heavy and light chains; 4) acquiring constant region sequences of the heavy and light chains; 5) preparing a full-length heavy chain vector; 6) co-transfecting a cell, taking the supernatant of cell culture, and purifying. The method utilizes different foot-and-mouth disease virus strains to infect cattle and obtains a neutralizing antibody capable of neutralizing three topotypes of serotype O FMDVs by screening, and thus can obtain a whole bovine-derived broadly neutralizing monoclonal antibody.

摘要: The present disclosure belongs to the technical field of biology, and in particular relates to an attenuated strain of African swine fever virus (ASFV) with IPTG-induced deletion of a D1133L gene and use thereof. In the present disclosure, it is firstly found that the D1133L protein of ASFV can inhibit production of IFN-β and downstream cytokines ISG-15 and ISG-56, and can be used as an immunosuppressant with a relatively strong immunosuppressive effect. In addition, the attenuated strain of ASFV with IPTG-induced deletion of the D1133L gene is constructed. Specifically, a screening expression cassette is inserted into a position before the non-structural protein gene D1133L of the ASFV using an Escherichia coli lac operator-repressor system, to obtain a recombinant virus. In the presence of the IPTG, the recombinant virus has similar characteristics to a wild-type virus; and in the absence of the IPTG, the expression of the D1133L protein is inhibited.
