    Abstract: The present invention is related to a retinoid-producing host cell, particularly oleaginous yeast, modified such that the percentage of retinyl acetate based on the total retinoids produced by such host cell is increased during fermentation using triglyceride oils, like for example vegetable oil, as carbon source, wherein the activity of certain endogenous hydrolases or transferases involved in undesired conversions of retinol or retinol acetate is reduced or abolished. Particularly, such modified host cell might be useful in a biotechnological process for production of vitamin A.

    Abstract: The present invention relates to a recombinant cell which is capable of producing one or more of itaconic acid, 4-methyl itaconate or 1-methyl itaconate, wherein the said recombinant microorganism has been modified in its genome such that it results in a deficiency in the production of a trans-aconitate methyltransferase. A recombinant yeast cell which is capable of producing itaconic acid and which overexpresses: —a nucleic acid encoding a polypeptide having cis-aconitate decarboxylase activity; and —one or more nucleic acids encoding polypeptides which separately or together catalyze a reaction towards acetyl CoA, wherein the said recombinant microorganism has been modified in its genome such that it results in a deficiency in the production of a trans-aconitate methyltransferase. These recombinant yeast cells may be used in processes for the production of itaconic acid, 4-methyl itaconate or 1-methyl itaconate.

    Abstract: The present invention relates to a method of identifying a heterologous polypeptide having enzymatic activity for converting pyruvate, acetaldehyde or acetate into acetyl-CoA in (the cytosol of) a yeast cell comprising: a) providing a mutated yeast cell comprising a deletion of at least one gene of the (PDH) by-pass, selected from the genes encoding the enzymes pyruvate decarboxylase (PDC), acetaldehyde dehydrogenase (ALD), and acetyl-CoA synthetase (ACS); b) transforming said mutated yeast cell with an expression vector comprising a heterologous nucleotide sequence encoding a candidate polypeptide having potential enzymatic activity for converting pyruvate, acetaldehyde or acetate into acetyl-CoA; c) testing said recombinant mutated yeast cell for its ability to grow on minimal medium containing glucose as sole carbon source, and d) identifying said candidate polypeptide as a heterologous polypeptide having enzymatic activity for converting pyruvate, acetaldehyde or acetate into acetyl-CoA in (the cytosol of) said yeast cell when growth of said cell is observed. The invention further relates to a method of producing a fermentation production such as butanol.
