    摘要: The present disclosure provides for a system and method for assessing chronic exposure of a biological sample, such as a bodily fluid, to an analyte of interest. A biological sample may be illuminated to thereby generate a one or more pluralities of interacted photons. These interacted photons may be detected to thereby generate one or more spectroscopic data sets representative of a biological sample. Spectroscopic data sets generated may be compared to at least one reference data set. Each reference data set may be associated with a known exposure to a known analyte. The present disclosure contemplates that the system and method disclosed herein may be used to analyze exposure of biological samples to at least one analyte over time. Data sets may be obtained at various time intervals to assess changes in a molecular composition as a result of chronic exposure to an analyte.

This arrangement of thin film layers may be such that it effectively enables a correction filter to generate a predetermined spectral response, wherein said predetermined spectral response is substantially the same as a determined instrument response correction associated with an instrument. The invention of the present disclosure therefore provides for effectively compensating for transmission inefficiencies associated with an instrument without the need for separate reference measurements to determine and correct for instrument response.

    摘要: A system and method to distinguish normal cells from apoptotic cells. A pre-determined vector space is selected where the vector space mathematically describes a first plurality of reference Raman data sets for normal cells and a second plurality of reference Raman data sets for apoptotic cells. A sample is irradiated with substantially monochromatic light generating a target Raman data set based on scattered photons. The target Raman data set is transformed into a vector space defined by the pre-determined vector space. A distribution of transformed data is analyzed in the pre-determined vector space. Based on the analysis, the sample is classified as containing normal cells, apoptotic cells, and a combination of normal and apoptotic cells. The sample includes the step of treating the sample with a pharmaceutical agent prior to irradiating the sample. Based on the classification, the therapeutic efficiency of the pharmaceutical agent is assessed.

    摘要: A system and method to automatically obtain spectra for samples. The method involves a two phase process including a photobleaching phase and a spectral acquisition phase. In the photobleaching phase, a series of spectral data sets of a sample are collected. A relative difference is determined between the background of subsequent spectral data sets is determined and compared to a predetermined threshold value. If threshold difference is less than the relative difference between the background of subsequent spectral data sets, the steps of collecting a series of spectra data sets is automatically repeated. In the spectrum acquisition phase, a series of Raman data sets of the sample are collected until a target SNR is obtained.

    摘要: A system and method for correction of instrument response of an optical spectroscopy instrument using a Raman standard sample supplied by NIST (National Institute of Standards and Technology). The smoother side of the NIST sample is placed facing a light collection optics in the spectroscopy instrument, whereas the non-smooth or rough side remains away from the light collection optics, but in contact with a platform or sample placement surface in the spectroscopy instrument. An instrument response function is determined with the NIST sample so placed. Thereafter, spectra or spectral images of target samples obtained using the spectroscopy instrument are divided by the instrument response function to correct for imperfections in the response of the optical spectroscopy instrument. The target sample spectra may be non-Raman spectra. The optical spectroscopy instrument may be a gratings-based or a tunable filter based chemical imaging system.
