Abstract: A trifunctional molecule is provided, comprising (i) a target-specific ligand, (ii) a ligand that binds a protein associated with a TCR complex, and (iii) a T cell receptor signaling domain polypeptide. Variants of the molecule are provided, including variants that exhibit optimized surface expression, transduction efficiency, and effector functionality. Variations include, for example, different ligands that bind CD3 epsilon (e.g., OKT3, L2K, F6A, UCHT1 and humanized UCHT1), different signaling domains, and different linkers between domains.

Abstract: The present application relates to compounds of Formula I comprising an antibody binding domain (ABD) comprising a hapten that binds to an antibody in a subject, the antibody comprising a hapten binding site, an antibody labelling domain (ALD) comprising a functional group that forms a covalent bond with an amino acid in the antibody that is proximal to the hapten binding site and the formation of the covalent bond results in elimination of the ABD and either a target binding domain (TBD) or a detection moiety domain (DMD), each domain being optionally connected with independently selected linkers. The present application also includes methods and uses of the compounds, for example, for immune recognition of target cells by recruited labelled antibodies.

Abstract: A packing assembly for forming a packed-bed for a chromatography device is described herein. The packing assembly includes a bottom plate and a first top plate. The first top plate has a top plate hole centrally positioned therein. The packing assembly also includes a middle plate. The middle plate has a middle plate hole centrally positioned therein. The middle plate hole is aligned with the top plate hole when the packing assembly is in an assembled state. The packing assembly also includes a second top plate. The second top plate has a protrusion extending outwardly. At least a portion of the protrusion has a same size and a same shape as the top plate hole to be received in the top plate hole when the packing assembly is in a packing state. Methods of packing chromatography media in a packing assembly and assembling a chromatography device are also described herein.

Abstract: The present application describes process for preparing an ortho-allylated hydroxy aryl compounds such as compounds of Formula (I) by reacting an allylic alcohol with a hydroxy aryl compound in the presence of aluminum compound selected from alumina and aluminum alkoxides and in a non-protic solvent wherein at least one carbon atom ortho to the hydroxy group in the hydroxy aryl compound is unsubstituted.
