The present invention provides a method for changing nitrogen utilization efficiency in a plant comprises regulating the expression of Arabidopsis NRT 1.7 or an ortholog thereof so that the nitrate remobilization from older leaves to young leaves in the plant is regulated, thereby the nitrogen utilization efficiency is changed. The present invention also provides a transgenic plant obtainable by transforming a plant with an expression construct with a high or low level of expression of NRT 1.7. On the other hand, the present invention yet provides a chimera nitrate transporter, a DNA molecule coding for this chimera transporter and an expression vector thereof.
Abstract:
This disclosure relates to a method to mimicking the germinal center reaction to generate antibodies with altered binding properties or increased affinity directly in hybridomas. To allow convenient and rapid affinity maturation of antibodies, a controllable activation-induced cytidine deaminase (AID) expression system to induce somatic hypermutation in hybridomas is developed. Selection of high affinity antibodies is achieved by fluorescence-activated cell sorting of hybridomas that preferentially bind fluorescence-labeled antigens. The disclosure also relates to de novo generation of hybridomas with tunable antibody affinity by generating myeloma fusion partners with controllable AID expression.

A sensor for detecting molecular interactions between a target and a binding domain by electrochemical impedance spectroscopy. The sensor includes an anodic aluminum oxide barrier layer having a gold-coated array of regularly spaced nano-hemispheres and gold nanoparticles coated with a binding domain attached thereto. Also provided are methods for producing the sensor and for using the sensor to detect the presence of a virus in a sample.

The present invention relates to a method for enhancing root growth of a plant by introducing a polynucleotide encoding late embryogenesis abundant protein, group 3 (LEA3) into the plant. Immunogenic compositions comprising partially glycosylated viral glycoproteins for use as vaccines against viruses are provided. Vaccines formulated using mono-, di-, or tri-glycosylated viral surface glycoproteins and polypeptides provide potent and broad protection against viruses, even across strains. Pharmaceutical compositions comprising monoglycosylated hemagglutinin polypeptides and vaccines generated therefrom and methods of their use for prophylaxis or treatment of viral infections are disclosed. Methods and compositions are disclosed for influenza virus HA, NA and M2, RSV proteins F, G and SH, Dengue virus glycoproteins M or E, hepatitis C virus glycoprotein E1 or E2 and HIV glycoproteins gp120 and gp41.

The present invention provides relates to a SKIN gene silencing plasmid comprising a promoter; two DNA fragments, which are obtained from one DNA fragment derived from the cDNA of SKIN1 or SKIN2 and arranged in sense and antisense orientation; and a third DNA fragment inserted between the two DNA fragments. SKIN is a negative regulator of the growth and yield of rice, and decreasing the expression of endogenous SKIN increases the yield of rice. The present invention also relates to a transformed plant cell and a transgenic plant comprising the SKIN gene silencing plasmid.

Crystal structure at 2.16 Å resolution of full-length Escherichia coli penicillin-binding protein 1b (PBP1b) in complex with its inhibitor moenomycin, is provided. 3D structures of amino acid residues involved in moenomycin binding and transglycosylation activity are identified. Binding sites for peptidoglycan synthesis inhibitors comprising amino acid residues from transglycosylase (TG), UvrB domain 2 homolog (UB2H) and transmembrane (TM) domains of PBP1b are identified at atomic level resolution. Rational drug design, based on the atomic coordinates, are disclosed. Methods for screening for antibiotics using anisotropic binding and transglycosylase inhibitor assays and novel antibiotics based on the screening assays are provided.
