    摘要: The invention concerns an infusion device for subcutaneous administering a medication or a therapeutic fluid to a patient. Said infusion device comprises a base element (1), which comprises fluid receiving means for receiving said fluid and fluid communication means for transferring said fluid into a cannula, said base element being provided with at least one recess (20) accommodating a septum (5) being pierceable by a needle. Said septum is secured to said base element by a septum housing (3) in such a way that a fluid transfer volume is formed in said at least one recess between an internal surface of said septum and an inner section of said recess. Said fluid transfer volume is communicating with said fluid communication means, and said septum housing is accommodating a premounted septum. The infusion device is characterized in that the septum housing is fixed and fluid sealed to said base element in said inner section of said recess.

    摘要: A metabolically engineered micro-organism has an operative first metabolic pathway in which a first metabolite is transformed into a second metabolite in a reaction in which NAD is a cofactor for a first enzyme, suitably a phosphorylating dehydrogenase, said reaction step producing NADH. Said second metabolite is transformed into at least one further metabolite in a reaction catalysed by a second enzyme, suitably a kinase. The organism has an operative second metabolic pathway characterised by an enzyme activity in excess of a native level in respect of a third enzyme, suitably a non-phosphorylating dehydrogenase, e.g. GAPN, catalysing a non-reversible reaction in which NADP is a cofactor and NADPH is a product. Said first metabolite is transformed into a said further metabolite without the involvement of said second enzyme.

    摘要: A recombinant microbial cell comprises at least one increased expressible enzyme activity controlling anabolic metabolism of ammonia as a nutrient source, the increased enzyme activity being an NADH-dependent activity catalysing the reaction (a) 2-oxoglutarate+NH3+NADH→glutamate+NAD or being an activity catalysing the reaction (b) 2-oxoglutarate+glutamine+NADH→2 glutamate+NAD or being an activity catalysing the reaction (c) glutamate+NH3+ATP→glutamine+ADP+Pi. The increased enzyme activity is encoded by a nucleic acid coding sequence linked to an expression signal not natively associated with the nucleic acid coding sequence and is increased as compared to the expression of the enzyme activity when the nucleic acid coding sequence is associated with its native expression signal.
