The present invention relates to a transdermal composition for dementia treatment containing donepezil as an active ingredient. The transdermal composition according to the present invention contains highly concentrated donepezil in a hydrophobic matrix, can continuously release the drug for a long time by having excellent long-term adhesion to the skin, and further exhibits consistently effective therapeutic effects over a long period of time by having a significantly improved skin penetration rate in comparison with conventional donepezil patches.

An aqueous composition comprises an amphiphilic block copolymer, having a hydrophilic block comprising pendant zwitterionic groups and a hydrophobic block, and a biologically active compound associated with the polymer. The polymer is preferably in the form of micelles, and preferably the biological active is a hydrophobic drug. The hydrophilic block is preferably formed from acrylic monomer including phosphorylcholine groups. The hydrophobic group is suitably formed from monomer which has groups which can be ionised at useful pH's, especially tertiary amine groups. Micelles may be formed by dissolving the block copolymer in aqueous solvent at a pH at which the amine groups are protonated then raising the pH to a value at which the amine groups are substantially deprotonated, whereupon micelles spontaneously form. The preformed micelles are then contacted with active, under conditions such that solubilisation of the active occurs. The active may be for tumour treatment.

The present invention relates to a biodegradable and thermosensitive poly(organophosphazene) with a functional group, a preparation method thereof, and a use thereof for delivery of bioactive substances. According to the present invention, poly(organophosphazene) is a phosphagen-based polymer showing biodegradability, thermosensitivity, and sol-gel phase transition depending on temperature change, whereby when administered into a living body with bioactive substances such as drugs, the poly(organophosphazene) forms a gel-phase at body temperature to be capable of controlled release of the bioactive substances. Further, the poly(organophosphazene) has functional groups to chemically bind with bioactive substances through an ionic bond, covalent bond, or coordinate covalent bond to be capable of a sustained release of the bioactive substances due to its good binding property. Therefore, the poly(organophosphazene) is useful as a delivery material for bioactive substances.

The present invention relates to poly(4-hydroxybutyrate)-b-monomethoxy(polyethylene glycol) copolymer nanoparticles, a method for preparing the same, and a pharmaceutical composition including the same as an active ingredient for treating brain disorders, in which the P(4HB)-b-mPEG nanoparticles according to the present invention break down slowly in brain cells such that gamma-hydroxybutyric acid (GHB) is released and so can sustainedly release a therapeutic level of concentration of GHB which is used as an agent for treating brain disorders such as epilepsy, and the nanoparticles are in the form of a hydrophobic polymer and so cannot easily be used as a hallucinogenic agent or narcotic agent due to the hydrophobic property whereby said nanoparticles do not dissolve in water or alcohol, and so can be used more safely than existing GHB, and hence, can advantageously be used instead of prior-art GHB therapeutic agents.
