Methods are provided for detecting single strand breaks in DNA sample comprising: contacting the DNA with a neutral hydroxylamine, denaturing the DNA and detecting single strand breaks in the denatured DNA. Methods are also provided for the detecting single strand breaks in DNA from a cell, the methods comprising lysing the cell, denaturing the DNA from the lysed cell, wherein the DNA is contacted with a neutral hydroxylamine prior to and/or during the denaturing step and detecting single strand breaks in the DNA from the cell. In addition, methods are provided for distinguishing between true single strand breaks and artifactual single strand breaks resulting from cleaved alkaline labile sites.

A moving image distribution system, wherein the user can view content moving images at no charge, and the provider of the content moving images can prevent a drain of the moving image contents and can protect the copy right thereof, as a copy right holder, without fail. The moving images are divided, along a time axis, into two master and slave nonreproducible moving image files. The slave moving image file (small file) is encrypted based on CM file data in which a predetermined CM element has been incorporated. In response to a reproduction request of a user terminal that possesses the master moving image file (main file) in advance, the slave moving image file is distributed with the CM moving images to the user terminal via a network line such as the Internet.

Succinic acid is produced by allowing a bacterium modified to enhance fumarate reductase activity or cell preparation thereof to react with an organic raw material in a reaction solution containing one of a carbonate ion, a bicarbonate ion, and carbon dioxide gas to generate succinic acid. More preferably, succinic acid is produced by allowing a bacterium modified to enhance activities of fumarate reductase and pyruvate carboxylase and decrease lactate dehydrogenase activity or cell preparation thereof to react with an organic raw material in a reaction solution containing one of a carbonate ion, a bicarbonate ion, and carbon dioxide gas to generate succinic acid. Succinic acid is obtained by collecting the produced succinic acid.

A method of producing coryneform bacteria having an improved amino acid- or nucleic acid-productivity comprises the steps of introducing a mutation in a promoter sequence of amino acid- or nucleic acid-biosynthesizing genes on a chromosome of a coryneform bacterium to make it close to a consensus sequence or introducing a change in a promoter sequence of amino acid- or nucleic acid-biosynthesizing genes on a chromosome of a coryneform bacterium by gene recombination to make it close to a consensus sequence, to obtain mutants of the coryneform amino acid d- or nucleic acid-producing microorganism, culturing the mutants and select a mutant capable of producing the intended amino acid or nucleic acid in a large amount. This method can construct a mutant capable of suitably enriching or controling the expression of an intended gene without using a plasmid and also capable of producing amino acids in a high yield, by the recombination or mutation.

L-arginine or L-lysine is produced by culturing a coryneform bacterium having an L-arginine- or L-lysine-producing ability and modified so that glutamine synthetase activity is enhanced, e.g., a coryneform bacterium which is modified so that adenylylation of glutamine synthetase is eliminated. L-arginine or L-lysine are produced by culturing the bacterium in a medium and allowing L-arginine or L-lysine to accumulate in the medium, and collecting the L-arginine or L-lysine from the medium.
