摘要:
A biological sterilization indicator, system, and methods of determining the effectiveness of a sterilization process. The biological sterilization indicator can include a locus of spores, a reservoir containing a liquid, and a sterilant path positioned to provide fluid communication between ambience and the locus of spores. The reservoir can have a closed state in which the reservoir is not in fluid communication with the locus of spores and an open state in which the reservoir is in fluid communication with the locus of spores. The biological sterilization indicator system can include the biological sterilization indicator and a detection device adapted to be coupled to the biological sterilization indicator. In some embodiments, the method can include assaying the spores for a detectable change in a characteristic, and detecting substantially all of the detectable change.

Compositions that comprise water, a first indicator reagent that can be converted by a first biological activity to a first biological derivative, and a plurality of particles are provided. The first indicator reagent can comprise a fluorogenic enzyme substrate having a fluorophore selected from the group consisting of umbelliferone, 7-aminocoumarin, β-naphthylamine, β-naphthol, fluorescein, resorufin, 9H-(1,3-dichloro-9,9-dimethyl acridin-2-one), rhodamine 110, a derivative of any of the foregoing fluorophores, and a combination of any of the foregoing fluorophores. The particles are capable of receiving and retaining the first biological derivative from an aqueous liquid. The first biological derivative can be indicative of a microorganism. The compositions further can comprise a gelling agent. Methods of using the compositions to detect the presence or absence of a microorganism in a sample are also provided.

The present invention provides method of detecting a predetermined biological activity. The method includes using an aqueous mixture comprising a first indicator reagent with a first absorption spectrum and a second indicator reagent. The second indicator reagent is converted by the predetermined biological activity to a second biological derivative with a second emission spectrum. The first absorbance spectrum includes detectable absorbance in at least a portion of wavelengths present in the second emission spectrum. The first indicator reagent is received and concentrated from an aqueous liquid by a substrate, facilitating the detection of the second biological derivative.

A system and method for preparing and analyzing samples. The system can include a sample preparation system and a sample detection system coupled to the sample preparation system. The sample preparation system can include a deformable self-supporting receptacle comprising a reservoir adapted to contain a liquid composition comprising a source and a diluent. The sample detection system can be positioned in fluid communication with the reservoir, and can be adapted to analyze a sample of the liquid composition for an analyte of interest. The system can further include a fluid path defined at least partially by the reservoir and the sample detection system. The method can include applying pressure to the deformable self-supporting receptacle to move a sample of the liquid composition in the fluid path to the sample detection system, and analyzing the sample for the analyte of interest with the sample detection system.

A method for partitioning an aqueous biological liquid sample into discrete microvolumes for detection and enumeration of microorganisms is described. The method involves distributing microvolumes of a sample to a plurality of hydrophilic liquid-retaining zones of a culture device, where each liquid-retaining zone is surrounded by a portion of a hydrophobic “land” area. Also disclosed are devices for carrying out these methods.

Method for detecting an analyte of interest in a sample. The method can include providing a container comprising a microstructured surface, and centrifuging the container toward the microstructured surface to form a sediment and a supernatant of the sample. Following centrifugation, the container can be inverted to decant at least a portion of the supernatant of the sample from the second portion, such that a concentrate (e.g., comprising the sediment) of the sample is retained in the microstructured surface. The concentrate can then be interrogated in the microstructured surface for the analyte of interest. In some embodiments, at least a portion of the second portion can be substantially transparent, such that the concentrate can be interrogated from the outside of the container, without requiring that the container be opened prior to interrogation.
