摘要:
The present invention provides a novel process for converting a standard inert amorphous calcium phosphate precipitate into highly reactive amorphous solids. The amorphous solids can be used to react with other calcium phosphate solids to form a poorly-crystalline synthetic hydroxyapatite that provides both bioactivity and structural integrity. This novel amorphous material can be reacted with other calcium phosphates at or below 37.degree. C. to form a bone-like material consisting of poorly crystalline hydroxyapatite.

A method of repair of patient tissues by implant including providing a living cell sample which is introduced into an implant member having a porous open structure. The cell sample may be cultured in the implant. The implant is secured to the patient, as by surgical implantation. In one embodiment, the implant portion which receives the cells preferably has a pore size of about 25 to 75 microns. In addition, a second pore size of about 100 to 400 microns for receipt of blood vessels and osteogenus cells through ingrowth after introduction into the patient, may be provided. The cell sample may advantageously be selected from the group consisting of cartilage cells, tendon cells, ligament cells and musculotendinous cells. The implant member may be advantageously used in bone or joint reconstruction surgery and in other forms such as artificial tooth implantation.A surgical implant comprising an inert member having, in one embodiment, a first series of open pores of an average size of about 25 to 75 microns and a second series of open pores of an average size of about 100 to 400 microns with patient cells growing within the pores.

In an aspect, a clamp system is provided including a clamp and a base, the base including a graft-engaging block including a graft-engaging surface engaging a first surface of the graft, and the clamp contacts the graft while at least a portion of the graft opposite the first surface is removed. In another aspect, a method of preparing a distal radius bone graft is provided including positioning the distal radius bone graft into a clamp such that at least part of a proximal portion and at least another portion of the bone graft are accessible, performing a depth cut on the proximal portion of the graft to form a prepared proximal face, and performing a width cut on the another portion to form a prepared face.

A tissue scaffold includes a first film having a plurality of cell openings and a second film adjacent the first film and having a plurality of cell openings larger than the cell openings of the first film. The cell openings of the first film interconnect with the cell openings of the second film to define pathways extending through the first and second films.

A shaped cartilage matrix isolated from a human or animal where the cartilage has been crafted to facilitate disinfection, cleaning, devitalization, recellularization, and/or integration after implantation. Also, a process for repairing a cartilage defect with the cartilage matrix. The matrix is in the form of an osteochondral plug including a cartilage cap ad subchondral bone, wherein one or more gaps, slats, bores, or channels extend through the tidemark at the interface between the cartilage cap and the subchondral bone.

Devices and methods for in situ drawing, filtering and seeding cells from the marrow of surrounding bone into a fusion cage without any of the challenges mentioned above. Various implants and devices with aspiration ports that enable in-situ harvesting and mixing of stem cells. These devices may include spinal fusion cages, long bone spacers, lateral grafts and joint replacement devices. Each device utilizes at least one aspiration port for harvesting of stem cell-containing marrow via aspiration from adjacent bony elements.

A system for harvesting bone graft material for use during bone fusion surgery. In one embodiment, a bone cutting tool has a blade fixed on a distal end of a shaft. The distal end of the shaft is pivoted on a cage set between two bones to be fused, and the blade is activated to cut into the bones and form a solid bone segment in each bone as the tool shaft turns. A paddle is arranged to be inserted between the bones, and to displace the cut bone segments so that a leading portion of each segment enters the bone opposite the bone from which the segment was cut, a central portion of the segment spans across the bones, and a trailing portion of the segment remains in the bone from which it was cut. The displaced segments act as strut grafts to fuse the bones to one another.

In some embodiments, the present invention provides tissue grafts, such as vascularized bone grafts, and methods for preparing and using such tissue grafts. In some embodiments the tissue grafts are made using pluripotent stem cells, such as autologous pluripotent stem cells. In some embodiments, the tissue grafts are made by creating a digital model of a tissue portion to be replaced or repaired, such as a bone defect, partitioning the model into two or more model segments, and then producing tissue graft segments having a size and shape corresponding to that of the model segments. Such tissue graft segments may be assembled to form a tissue graft having a size and shape corresponding to that of the tissue portion to be replaced or repaired.

A device for perfusing porous biomaterials with biological liquids includes a tubular body with an axial cavity defining a perfusion chamber for containing a porous biocompatible material, a connector at a distal end of the tubular body for receiving a perfusion liquid, a plunger having a distal end slidably movable in the cavity to withdraw air from the biomaterial, a vent for releasing the air withdrawn from the biomaterial and contained in the chamber, and a filter permitting selective passage of the air contained in the chamber toward the vent, while preventing leakage of the liquid to the outside. An air collector communicates with the chamber and the vent, and a control unit is associated with the plunger and designed to be actuated by the user to control the air flow from the air collector to the vent.A method of perfusing porous biomaterials with biological liquids.
