    摘要: A system for harvesting bone graft material for use during bone fusion surgery. In one embodiment, a bone cutting tool has a blade fixed on a distal end of a shaft. The distal end of the shaft is pivoted on a cage set between two bones to be fused, and the blade is activated to cut into the bones and form a solid bone segment in each bone as the tool shaft turns. A paddle is arranged to be inserted between the bones, and to displace the cut bone segments so that a leading portion of each segment enters the bone opposite the bone from which the segment was cut, a central portion of the segment spans across the bones, and a trailing portion of the segment remains in the bone from which it was cut. The displaced segments act as strut grafts to fuse the bones to one another.

    摘要: The invention provides methods and compositions for differentiating stromal cells from adipose tissue into cells having osteoblastic properties, and methods for improving a subject's bone structure. The methods comprise culturing stromal cells from adipose tissue in β-glycerophosphate and ascorbic acid and/or ascorbate-2-phosphate for a time sufficient to allow differentiation of said cells into osteoblasts. Such methods and compositions are useful in the production of osteoblasts for autologous transplantation into bone at a surgical site or injury. The compositions comprise adipose stromal cells, a medium capable of supporting the growth of fibroblasts and amounts of β-glycerophosphate and ascorbic acid and/or ascorbic-2 phosphate sufficient to induce the differentiation of said stromal cells into osteoblasts.The invention further provides methods of identifying compounds that affect osteoblast differentiation. Such compounds are useful in the study of bone development and in the treatment of bone disorders, including bone fractures and osteoporosis.

    摘要: The present invention provides a novel process for producing a calcium phosphate cement or filler which hardens in a temperature dependent fashion in association with an endothermic reaction. In the reaction a limited amount of water is mixed with dry calcium phosphate precursors to produce a hydrated precursor paste. Hardening of the paste occurs rapidly at body temperature and is accompanied by the conversion of one or more of the reactants to poorly crystalline apatitic calcium phosphate. The hardened cements, fillers, growth matrices, orthopedic and delivery devices of the invention are rapidly resorbable and stimulate hard tissue growth and healing. A composite material is provided including a strongly bioresorbable, poorly crystalline apatitic calcium phosphate composite and a supplementary material. The supplementary material is in intimate contact with the hydroxyapatite material in an amount effective to impart a selected characteristic to the composite. The supplemental material may be biocompatible, bioresorbable or non-resorbable. A method for treating a bone defect also is provided by identifying a bone site suitable for receiving an implant, and introducing a strongly resorbable, poorly crystalline apatitic calcium phosphate at the implant site, whereby bone is formed at the implant site. The implant site may be a variety of sites, such as a tooth socket, non-union bone, bone prosthesis, an osteoporotic bone, an intervertebral space, an alveolar ridge or a bone fracture.

    摘要: The invention is directed towards a process for implanting a cartilage graft into a cartilage defect and sealing the implanted cartilage graft with recipient tissue by creating a first bore down to the bone portion of the cartilage defect, creating a second shaped bore that is concentric to and on top of the first bore to match the shape and size of the cartilage graft, treating the first bore and the second shaped bore at the defect site with a bonding agent, treating the circumferential area of the cartilage graft with a bonding agent, inserting the cartilage graft into the defect site and wherein the superficial surface of the cartilage graft is at the same height as the surrounding cartilage surface. The first and second bonding agents may be activated by applying a stimulation agent to induce sealing, integration, and restoration of the hydrodynamic environments of the recipient tissue. The invention is also directed towards a process for repairing a cartilage defect and implanting a cartilage graft into a human or animal by crafting a cartilage matrix into individual grafts, cleaning and disinfecting the cartilage graft, applying a pretreatment solution to the cartilage graft, removing cellular debris using an extracting solution to produce a devitalized cartilage graft, implanting the cartilage graft into the cartilage defect with or without an insertion device, and sealing the implanted cartilage graft with recipient tissue. The devitalized cartilage graft is optionally recellularized in vitro, in vivo, or in situ with viable cells to render the tissue vital before or after the implantation. The devitalized cartilage graft is also optionally stored between the removing cellular debris and the recellularizing steps. The invention is further directed toward a repaired cartilage defect.
