Selection of clones having the kinase and/or phosphatase-like structure from clones which had been isolated and the structures thereof had been determined in the Helix Research Institute (helix clones; Japanese Patent Application No. 2000-183767) was conducted. Two novel genes were provided by carrying out homology search for all the helix clones by using the amino acid sequences of known kinases and phosphatases as queries. The genes are expected to be involved in intracellular signal transduction. The physiological functions of the inventive genes can be tested by using reporter gene assay systems capable of detecting signal transduction. The proteins of the present invention are useful as target molecules in drug discovery and in the development of new pharmaceuticals.

The electrophoresis apparatus includes plural capillaries for separating fluorephore-labeled samples by electrophoresis, fluorescence detecting parts provided in a part of these capillaries arranged in the same place for detecting a fluorescence emitted by fluorephore labels when a part of the plural capillaries is scanned and irradiated by a laser beam, and a fluorescence detection system for detecting this fluorescence. The fluorescence detecting parts are scanned and repeatedly irradiated by the laser bean where a scanning period of the fluorescence detecting parts by the laser bean is t1, and the fluorescence is detected by the fluorescence detecting system where an acquisition time of fluorescence signal is t2 (t1≦t2). The laser bean from a laser source is narrowly converged by a light collecting lens, and a galvanomirror is rotated in a rotation directional of the galvanomirror around the rotation axis of the galvanomirror so as to repeatedly scan the fluorescence detecting parts.

A multicolor fluorescence detection type electrophoresis apparatus comprises an electrophoretic device having migration lanes in which fragment groups specifically labeled by fluorophores of different light emission peak wavelengths are mixed, added, and migrated. It identifies and detects the fragment groups. It uses laser beams of different wavelengths for exciting the fluorophores in the migration lanes. The laser beams are irradiated to separate positions on the migration lanes for detection of a few kinds of fluorophores at irradiation positions. For the purpose, multicolor optical filters are arranged near the irradiation positions to identify and detect lights emitted from the fluorophores excited by wavelength at every irradiation position.

In a DNA detecting method wherein the fluorescence detection type electrophoresis apparatus is used, Texas Red is employed as a fluorophor which labels a DNA fragment, the He-Ne laser having an emission wavelength of 594 nm is used as an excitation light, the excitation light irradiates the line portion of the gel plate simultaneously, and a resulting linear fluorescent image is collected by a cylindrical lens and photodetected, thereby providing an apparatus featuring a higher sensitivity, a smaller size and a lighter weight than conventional apparatuses.

An electrophoretic apparatus includes an electrophoresis panel having a fluted glass plate and a non-fluted plane plate. The fluted glass plate is provided with a plurality of substantially parallel narrow electrophoresis grooves and a groove intersecting the electrophoresis grooves for laser beam irradiation. The fluted glass plate and the non-fluted plane plate are closely superposed on each other to form many capillaries adapted to be filled with gel to thereby form many electrophoresis lanes. The electrophoretic apparatus further includes a light measuring instrument to measure fluorescence images at positions which are subjected to laser beam irradiation. Passage of fluorescently labeled sampled fragments through the laser beam irradiation positions in each electrophoresis lane is detected by measuring a change of fluorescence emission with time.

A cDNA fragment participating in the maintenance of smooth muscle differentiation was isolated using a culture system of chicken gizzard smooth muscle cells, the differential display method and the subtracted hybridization method. Using the obtained cDNA sequence as a query, cDNA sequences of Helix Research Institute (Japanese Patent Application No. 2000-118776) were retrieved, and thus, a novel gene “C-NT2RP3001495” was obtained. The protein encoded by this gene has two WW domains that participate in protein interactions in the N-terminal domain. Evidence suggests that this protein binds to other proteins, and thus regulates the intracellular signal transduction, gene expression, and so on, thereby participating in the maintenance of the differentiation of smooth muscle cells. This protein and compounds regulating the expression thereof are markedly useful in developing drugs for various diseases associated with abnormality in the maintenance of smooth muscle cell differentiation.

The full-length nucleotide sequences of the cDNA and amino acid sequences encoded by the nucleotide sequences have been determined. Because the cDNA of the present invention are full-length and contain the translation start site, they provide information useful for analyzing the functions of the polypeptide.

In the multi-color fluorescence detection type electrophoresis apparatus provided with the electrophoresis gel plate, excitation laser source, means to separate the fluorescence images according to each emission wavelength, and the detector of the fluorescence subjected to wavelength selection, two or more laser sources are provided, each of laser lights is irradiated on the sample on a time-sharing basis, and the filter which cuts off the scattered light of each the laser synchronous with the laser beam is installed in front of the wavelength separation means thereby providing simultaneous, quick and real-time analysis of a great number of samples such as DNA and RNA labeled by many types of fluorophores, without overlapping the wavelengths of the excitation light and the fluorescence.

Determination of a plurality of species of antibodies or antigens is attained by a method which comprises forming a plurality of different kinds of reaction membranes each having a different species of antibody or antigen on an electrophoretic carrier, superposing these reaction membranes, optionally superposing a filter on the laminate of reaction membranes, inserting the laminate in an electrolyte, adding a plurality of different species of antigens or antibodies corresponding to the plurality of species of antibodies or antigens supported in the aforementioned reaction membranes, electrophoretically moving the added antigens or antibodies through the electrolyte and enabling them to react with the antibodies or antigens supported on the reaction membranes, and measuring the concentrations of either the antigens or antibodies resulting from the reaction or the antibodies or antigens supported in an unreacted form on the reaction membranes.
