Abstract: Methods of maintaining narrow residence time distributions in continuous flow systems, particularly applicable to virus inactivation such as during a protein purification process. Fluid sample is introduced into an axial flow channel and caused to flow therein in discrete packets or zones to minimize residence time distribution and axial dispersion. Embodiments described herein obviate or minimize the need for using large tanks or reservoirs for performing virus inactivation during a protein purification process; reduce the overall time required for virus inactivation, and/or reduce the overall physical space required to perform the virus inactivation operation during a protein purification process, which in turn reduces the overall footprint for the purification process.

Abstract: A process for the primary clarification of feeds, including chemically treated flocculated feeds, containing the target biomolecules of interest such as mAbs, mammalian cell cultures, or bacterial cell cultures, using a primary clarification depth filtration device without the use of a primary clarification centrifugation step or a primary clarification tangential flow microfiltration step. The primary clarification depth filtration device contains a porous depth filter having graded porous layers of varying pore ratings. The primary clarification depth filtration device filters fluid feeds, including chemically treated flocculated feeds containing flocculated cellular debris and colloidal particulates having a particle size distribution of approximately about 0.5 μm to 200 μm, at a flow rate of about 10 litres/m2/hr to about 100 litres/m2/hr. Kits and methods of using and making the same are also provided.

Abstract: A depth filtration device for the clarification of biological fluids including a composite depth filter media having a nonwoven first layer integral with a second layer containing a polyacrylonitrile (PAN) fibers, a filter aid, and a wet-strength resin. The depth filter media exhibits increased binding capacity for soluble impurities such as DNA and host cell proteins from biological/cell culture feedstreams during secondary clarification and low-level impurity clearance of harvested cell culture fluids, such as those used for the manufacture of monoclonal antibodies. The depth filter media additionally exhibits significantly lower flushing requirements, resulting in lower levels of organic, inorganic and bioburden extractables released, high dirt holding capacities and good chemical and/or radiation resistance.

Abstract: A clamp including a first jaw member having a camming surface, a second jaw member pivotable with respect to the first jaw member, a first biasing member biasing the first and second jaw members to a first position; and a locking mechanism that includes a cam or roller for cooperating with the camming surface of the jaw member, and a second biasing member biasing the cam or roller against the camming surface to lock the first and second jaw members in a second position. An optional alignment collar can be used to align components of the filtration apparatus prior to clamping. Filtration apparatus including the clamp, and also optionally including the alignment collar, is also disclosed.

The valve further comprises an extended flange having a surface that is approximately coplanar with a surface of the second position when the plunger is displaced toward the proximal end of the bore, creating a zero dead leg position

Abstract: A system, software application and method that allows a customer to protect their proprietary database of compounds and substances while utilizing a retrosynthesis software application is disclosed. The customer's proprietary database is encrypted prior to being provided to the retrosynthesis system. This encrypted is performed using a hash and optionally a salt. The retrosynthesis algorithm then creates synthons as is traditionally done. However, after their creation, the synthons are hashed so that they may be compared to the entries in the customer's proprietary database. In this way, the actual contents of the customer's database are never made available to the retrosynthesis system or software application.
