Abstract: A sensing device applied to an analyte molecule of a liquid sample and a buffer flow has at least one first inlet, at least one second inlet, a micro-flow channel, and at least one immobilization element. The first inlet is for inputting the liquid sample. The second inlet is for inputting the buffer flow. The micro-flow channel communicates with the first inlet and the second inlet. The immobilization element is for immobilizing sensing molecules for the analyte molecules. The analyte sample flow and the buffer flow, in the reverse direction, in the micro-flow channel enable the association and disassociation kinetics to be obtained. The present invention further provides a sensing system and a sensing method using the sensing device.

Abstract: Methods of assaying a biological target are disclosed. The method comprises: (a) providing a sample containing the biological target; (b) providing biotin-labeled first aptamers conjugated to a gold nanoparticle (GNP), and second aptamers conjugated to a magnetic bead, wherein the first and the second aptamers exhibit specific binding affinities to the target; (c) incubating the sample with the first and the second aptamers to obtain target-bound aptamers; (d) separating the target-bound aptamers from unbound aptamers; (e) eluting the first aptamers from the GNP; (f) incubating the eluted biotin-labeled first aptamers with streptavidin-magnetic beads and reporter gold nanoparticles (GNPs) to obtain a complex comprising: the bead, the first aptamers, attached to the bead; and the reporter GNPs captured by the bead through the first aptamers; (g) eluting the reporter GNPs captured; and (h) detecting the target by measuring and analyzing a light-scattering signal of the eluted reporter GNPs.

Abstract: The crystal structure at 2.16 Å resolution of the full-length bacterial bifunctional transglycosylase penicillin-binding protein 1b (PBP1b) from Escherichia coli, in complex with its inhibitor moenomycin, is provided. The atomic coordinates of the complex as well as the moenomycin binding site are provided. Three dimensional structures of amino acid residues involved in moenomycin binding and transglycosylation activity are identified. Binding site for peptidoglycan synthesis inhibitors comprising inhibitor-binding site comprises amino acid residues from at least one of transglycosylase (TG), UvrB domain 2 homolog (UB2H) and transmembrane (TM) domains of PBP1b are identified at an atomic level of resolution. Methods for rational drug design based on the atomic coordinates are provided. Methods for screening for antibiotics based on anisotropic binding assay and transglycosylase inhibitor assays are provided. Novel antibiotics based on the screening assays of the invention are disclosed.
