    摘要: A multi-layered tissue construct includes: a first layer comprising a first hydrogel; and a second layer comprising a second hydrogel, wherein the first layer is connected to the second layer at a first transition zone and wherein at least one of the first layer and the second layer further comprises a component selected from the group consisting of cells and a bioactive substance. Another multi-layered tissue construct includes: a first layer comprising a first hydrogel; a second layer comprising cells of a first type, wherein the second layer is disposed on the first layer; and a third layer comprising a second hydrogel and optionally cells of the first type encapsulated in the second hydrogel, wherein the third layer is disposed on the second layer. Methods for producing these multi-layered tissue constructs are also disclosed.

    摘要: Compositions for tissue engineering and drug delivery have been developed based on solutions of two or more polymers which form semi-interpenetrating or interpenetrating polymer networks upon exposure to active species following injection at a site in a patient in need thereof. The polymers crosslink to themselves but not to each other; semi-interpenetrating networks are formed when only one of the polymers crosslink. The resulting viscous solutions retain the biologically active molecules or cells at the site of injection until release or tissue formation, respectfully, occurs.As a result of studies conducted with polymer-cell suspensions forming interpenetrating polymer networks, it has been determined that polymer solutions can be formulated wherein the active species is provided by exposure of the polymer solution to an exogenous source of active species, typically electromagnetic radiation, preferably light. Studies demonstrate that light will penetrate through skin, body fluids (such as synovial fluid) and membranes and polymerize the polymer solutions. The polymer solutions can be crosslinked ionically or covalently, to form a hydrogel, semi-interpenetrating polymer network or an interpenetrating polymer network.

    摘要: The present invention relates to the covalent binding of a hydrogel to an extracellular matrix (ECM). The integration of the hydrogel with the tissue is superior to that in previous techniques. Moreover, unlike previous techniques, the present invention does not require a photoinitiator. Potential therapeutic applications include tissue repair and delivery of drugs or cells.The ECM is first exposed, then treated with a priming agent. Then a polymerizable agent is added and crosslinked to the ECM.Two primary embodiments of methods are disclosed. In the first, the priming agent is an oxidizer which creates tyrosyl radicals in the ECM, which are then bound by acrylate groups in the polymerizable agent. In the second, the priming agent contains aldehydes which bind amino groups in the ECM.
