Methods are provided for analyzing characteristics of fluids in the context of an acoustic ejection system. Such a system has a controller, an acoustic radiation generator, and a coupling medium coupling the radiation to a reservoir holding fluid. The methods can use acoustic radiation to both perturb a surface of the fluid in the reservoir and analyze the effect of the perturbation. The methods may use information about prior fluids. The methods of the invention can determine physical characteristics such as speed of sound and viscosity. The methods also include ways to determine a level of acoustic energy suitable to eject a droplet. Preferably the methods are executed automatically under control of programming of a controller of an acoustic ejection system.

A printing method and apparatus is disclosed wherein the printhead has a linear array of nozzles oriented in an axis, and drops of a liquid are printed at a particular location from first and second nozzles of the printhead at a first location. In an embodiment, the nozzles are arranged in groups, each group of nozzles being used to superpose spots of a particular liquid at a different location. A further method is described wherein the printhead is triggered by transferring printing triggers from a processing module to a printhead at print intervals representative of a plurality of fixed units of spatial displacement of the printhead. The present invention is particularly advantageous for the cycling of print nozzles, superposition of spots in linear arrays and fine control of spot positioning. The invention has particular application in the field of printing DNA microarrays.

Acoustic waves are used to transfer small amounts of fluid in a non-contact manner. Acoustic waves are propagated through a pool of a source fluid in such a manner that causes the ejection of a single micro-droplet from the surface of the pool. The droplet is ejected towards a target with sufficient force to provide for contact of the droplet with the target. Because the fluid is not contacted by any fluid transfer device such as a pipette, the opportunities for contamination are minimized. Methods may be employed to transfer fluids from an array of source sites to an array of target sites, thereby enabling the precise automation of a wide variety of procedures including screening and synthesis procedures commonly used in biotechnology.
