The antibody is obtained by immunizing an animal with an immunogen containing parathyroid hormone or fragment thereof oxidized at methionines at positions 8, 18 or both; and a recovering of antibodies; whereby the complementary determining region of the antibody or antibody fragment or single chain antibody specifically recognizes a conformational epitope (antigenic determinant) which is a tertiary structure and only present on oxidized parathyroid hormone and fragments thereof only but not regular bioactive human parathyroid hormone, while the binding surface to the complementary binding region does not include any one of the oxidized methionines of human parathyroid hormone.

Antibodies for a variant of intact parathyroid hormone (PTH) or its fragments which have at position 17 the amino acid serine which is phosphorylated [p(17) PTH] and methods of producing the same. The invention further includes various immunoassay methods which use the described antibody alone or in combination with other antibodies to determine the circulating levels of p(17) intact PTH (1-84) or fragments thereof in biological fluids. Such antibodies and immunoassays can be used in conjunction with other antibodies to detect all biologically active forms of hPTH present in biological fluids.

The present invention discloses the use of PSP-94 for treating a patient suffering from hypercalcemia of malignancy. Prostate secretory protein of 94 amino acids (PSP-94) is known to serve as prognostic marker in hormone-dependent prostate. In the current study the effect of PSP-94 on prostate cancer growth, metastases to the skeleton and hypercalcemia of malignancy was examined. A marked dose-dependent decrease in primary tumor volume was seen in experimental animals receiving PSP-94. Furthermore, while control animals routinely developed hypercalcemia due to PTHrP production, treatment with PSP-94 led to a near normalization of plasma calcium and a marked reduction in PTHrP production as determined by radioimmunoassay and immunohistochemistry. Results obtained demonstrate the ability of PSP-94 to be an effective treatment modality for prostate cancer. Furthermore, decrease in plasma PTHrP and calcium levels can serve as useful biochemical markers for monitoring the efficacy of this novel anti-tumor agent.
