摘要: It is considered that the activation of pro-MMP-2 by MT1-MMP is one of the important steps of tumor cell infiltration such as cancer. However, the detailed mechanism of the expression of this activity still remains unknown. It is therefore required to clarify the mechanism of the activation of pro-MMP-2 by MT1-MMP and thus clarify not only the infiltration and metastasis of cancer but also various phenomena in which MT1-MMP seemingly participates, thereby contributing to the prevention and treatment of various diseases. In the activation of pro-MMP-2 by MT1-MMP, it is essentially required to form complexes consisting of plural MT1-MMPs, in particular, an MT1-MMP homodimer. The formation of this dimer is based on the function of the PEX region in MT1-MMP. By using these findings, it is intended to inhibit the formation of the MT1-MMP homodimer and inhibit the activation of pro-MMP-2, thereby regulating the infiltration and metastasis of cancer, etc.

摘要: Drugs characterized by containing hydroxamic acid derivatives as the active ingredient which are efficacious in treating and/or preventing allergies, in particular, type I and/or type II allergies, etc. These drugs exert therapeutic and/or preventive effects on inflammation, rhinitis, conjunctivitis, bronchial asthma, atopic dermatitis (dermatitis, enteritis, etc.) and allergic digestive inflammation. Use of these drugs achieves the effects of: (A) inhibiting the proliferation of colonies of blood cells (lymphocytes, etc.) in an affected part; and/or (B) relieving inflammation caused by the migration, infiltration, accumulation, etc. of blood cells (lymphocytes, etc.) into an affected part; and/or (C) regulating the pathophysiological functions of cells such as blood cells (lymphocytes, etc.), Langerhans cells and dendritic cells; and/or (D) regulating the production of antibodies, in particular, IgE in the plasma, thus being useful in treating and/or preventing diseases or pathological conditions in the affected parts.

摘要: A shuttle vector is constructed by preparing a DNA region replicable in a microorganism belonging to the genus Rhodococcus from a Rhodococcus-origin plasmid having the base sequence represented by SEQ ID NO:73 and a plasmid having the base sequence represented by SEQ ID NO:74, DNA fragments thereof or the like and further preparing a DNA region replicable in Escherichia coli from an E. coli-origin plasmid or its DNA fragment. Then an aminoketone asymmetric reductase gene is inserted into the shuttle vector to form a vector. Next, a transformant having this vector is constructed to thereby produce aminoketone asymmetric reductase and an optically active aminoalcohol.

摘要: It has been required to economically produce a large amount of an optically active gamma-lactone derivative (for example, pantolactone) which is useful as an intermediate in synthesizing a useful substance such as a drug. To achieve this object, it is advantageous to employ a technique of enzymatic asymmetric hydrolysis with the use of an enzyme lactonase. However, it remains troublesome to prepare the enzyme or utilize a microorganism capable of producing the enzyme. Also, it is difficult to obtain a sufficient and stable enzymatic activity in the case of using recombination techniques. In producing lactonase, which asymmetrically hydrolyzes a gamma-lactone derivative such as racemic pantolactone selectively, by a recombination technique, maturation lactonase DNA and signal peptide region DNA are both transferred into a host. Thus, a stable activity comparable to that of natural lactonase can be achieved and a transformant having a high enzyme activity and sustaining it in a stable state can be obtained. As a result, it becomes possible to efficiently and industrially advantageously perform the asymmetric synthesis of a gamma-lactone derivative.
