Abstract: Novel compositions for removing impurities such as, protein aggregates, from a sample containing a protein of interest, e.g., an antibody. Such compositions can be used prior to the virus filtration step during protein purification, to remove aggregates and protect the virus filter from fouling, therefore improving virus filter capacity. A porous solid support including a co-polymer having at least two monomers, wherein at least one of the monomers comprises acrylamide and at least a second monomer comprises a hydrophobic binding group, where the solid support selectively binds protein aggregates, thereby to separate the monomeric protein of interest from the protein aggregates. The method can be performed under neutral to high pH and high conductivity conditions.

Abstract: Chromatography media including a high surface area thermoplastic porous nanofiber and an ion-exchange ligand functionality on the surface of the fiber. The porous nanofibers display a convoluted structure that is comprised of discrete bundles of highly entangled nanofibrils that may be fibrillated or ridged. The porous fibers can be prepared through the extraction of a dissolvable mineral or polymeric porogen that is embedded into the fiber during its manufacture in a melt extrusion process.

Abstract: Methods of filtering a liquid feed are disclosed. In one version, the method comprises passing a liquid feed through a single pass tangential flow filtration (SPTFF) system and recovering the retentate and permeate from the system in separate containers without recirculation through the SPTFF system. In another version, the method of filtering a liquid feed, comprises passing a liquid feed through a tangential flow filtration (TFF) system, recovering permeate and a portion of the retentate from the system in separate containers without recirculation through the TFF system, and recirculating the remainder of the retentate through the TFF system at least once. The methods can be performed using an SPTFF or a TFF system that comprises manifold segments to serialize the flow path of the feed and retentate without requiring diverter plates.

Abstract: A testing apparatus employs an interface module removably attached to a filter testing device. The interface module includes a first flowpath communicating with an outlet of the testing device and a housing of the filter device, a first valve disposed in the first flowpath, a second flowpath communicating with the housing of the filter device and an exhaust, a second valve disposed in the second flowpath, and an enclosure enclosing the first flowpath, the first valve, the second flowpath, and the second valve. The interface module is cleanable and/or sterilizable, and replaceable. A filter integrity testing method uses an interface module to prevent contamination of the testing device and cross-contamination of different filter devices and avoid liquid ingress into the testing device.

Abstract: The present invention provides a filter element (500) having a radial permeate discharge path (550). The filter element generally includes a closed membrane structure (510) wrapped about a core (530) in reciprocating clockwise and counterclockwise directions, forming semicircular folds of membrane about the core. The semicircular folds of membrane have opposingly situated apical ends (560) separated by a gap. The interior of the closed membrane structure (510) defines a feed channel and the exterior of the closed membrane structure defines at least one permeate channel (522). A radial permeate discharge path (550) extends through the gap between the apical ends of the semicircular folds of membrane. Systems containing, and methods of using, filter elements including radial permeate discharge paths are also provided.

Abstract: The present invention provides a process for incorporating a wireless device into a plastic container as part of the steps of manufacturing the container. The plastic container can be a flexible plastic bag and are formed of one or more sheets, generally two or more sheets of plastic film or a rigid plastic container such as a tub or tote. The bags are formed by sealing together adjacent edge portions of each film layer. One embodiment of the present invention is to incorporate a wireless device between the edge portions of the film(s) before or during sealing so that the wireless device becomes permanently sealed into the film material but is isolated from both the bag interior and the outside environment. Another embodiment is to incorporate the wireless device onto or into a plastic component that is sealed to the container, such as a nipple or port, so that the wireless device becomes permanently sealed to the container but is isolated from both the container interior and the outside environment.

Abstract: A method of aerosol integrity testing of filters, capable of detecting single defects that are less than 20 μm in diameter, and even as small as 2 μm in diameter, in liquid sterilizing grade filters such as filter cartridges. The method challenges the filter in a dry state with a particle stream of aerosol particles of the appropriate size and in the appropriate concentration, such that at least one or more of the particles in the stream will penetrate a defective region or regions within the membrane but will not penetrate in the integral region of the membrane. Wetting of the filter is not required.
