The subject invention pertains to methods and compositions for protecting feline animals from infection by FIV using immunogens derived from primate immunodeficiency viruses, including HIV and SIV. Methods for vaccinating feline animals with the subject vaccine compositions are described. Feline animals vaccinated according to the methods and compositions of the subject invention exhibit protective humoral and cellular immune responses to FIV when challenged with FIV.

We have developed DNA and viral vectors that can be used, alone or in combination, as a vaccine against one HIV lade, subtype, or recombinant form of HIV or against multiple HIV clades, subtypes, or recombinant forms. Moreover, the vectors can encode a variety of antigens, which may be obtained from one clade or from two or more different clades, and the antigens selected and/or the manner in which the vectors are formulated (e.g., mixed) can be manipulated to generate a protective immune response against a variety of clades (e.g., the clades to which a patient is most likely to be exposed; with the proportions of the components of the vaccine tailored to the extent of the patient's risk to a particular dade or clades).

Disclosed is a novel HIV gene comprising a set of open reading frames encoded with the template as the plus strand of the proviral DNA, and located in the region of HIV-1 long terminal repeat. The genes encode a set of antisense proteins, (HAPs) as well as smaller proteins, related to, and containing structural motif resembling that of chemokine proteins. Depending upon the ribosomal frameshift, a plurality of proteins may be translated from the antisense RNA. The smaller proteins have similarity with chemokine SDF-1 and may play a role as a cofactor with gp120 in the binding to and entry of HIV to a target cell.

The molecules have a first nucleic acid sequence encoding in antigen containing two or more cytolytic T lymphocyte (CTL) epitopes, wherein said epitopes are selected from the amino acid sequences of SEQ ID NOs: 1, 2, 3, 4, 5 and 6 and analogues of any thereof which can be recognised by a CDS8null T cell that recognises an epitope with the amino acid sequence of any one of SEQ ID NOs: 1, 2, 3, 4, 5 or 6. Peptides encoded by the molecules and vectors and compositions containing these molecules are also described. Methods of eliciting an immune response using these molecules are also described.

The present invention provides a population of live attenuated recombinant replication-competent viruses, which population comprises at least two member viruses, each of the member viruses comprising a nucleotide sequence encoding a different antigenic polypeptide from a source other than a parent virus from which the recombinant virus was derived. When a eukaryotic cell of a mammalian host is infected with a member of the population, the nucleotide sequence is expressed, the antigenic polypeptide is produced, and elicits an immune response. The invention further provides compositions, including immunogenic compositions, comprising a subject virus population. The invention further provides methods of eliciting an immune response to a polypeptide in an individual, involving administering a subject virus population. The invention further provides devices for use in eliciting an immune response.
