A method comprises the steps of providing a first mold with a high and low features. A first layer is formed over the features. The high feature extends a predetermined height through the first layer to define a via or extends near to the first layer to define a membrane of predetermined thickness. The low feature defines a lower channel in the first layer which is communicated with the via or membrane. The second layer has an upper channel formed therein, so that the high feature extends into the upper channel in the second layer or is positioned adjacent to the upper channel in the second layer. The first mold is removed. The partially completed structure is assembled onto a substrate to result in a via, septum or microfuse formed between different, adjacent vertical levels in the multilayer microfluidic circuit.

The means for dependent on flow for creating a pressure differential comprises either a dead-end detour channel from the flow channel to the valve, or a loop channel fed back to the control chamber of the valve.

These devices enhance and miniaturize microfluidic control, thereby expanding the available capabilities and allowing complete system miniaturization for handheld diagnostic apparatuses.

Microfluidic assay detectors and microfluidic assay detection methods are disclosed. A microfluidic chip is coupled to a light emitting device, emission filters and excitation filters. Excited fluorescent light is detected by a camera and a lens. The correspondent reading allows parallel detection of features such as antigens and biomarkers. A microfluidic filter and related methods are also disclosed. The filter can be used with on-chip fluid filtration such as whole blood filtration for microfluidic blood analysis. The filter is able to filter the necessary volume of fluid and in particular blood in an acceptable time frame.

The development of a high-throughput multi-antigen microfluidic fluorescence immunoassay system is illustrated in a 100-chamber PDMS (polydimethylsiloxane) chip which performs up to 5 tests for each of 10 samples. Specificity of detection is demonstrated and calibration curves produced for C-Reactive Protein (CRP), Prostate Specific Antigen (PSA), ferritin, and Vascular Endothelial Growth Factor (VEGF). The measurements show sensitivity at and below levels that are significant in current clinical laboratory practice (with SIN>8 at as low as 10 pM antigen concentration). The chip uses 100 nL per sample for all four tests and provides an improved instrument for use in scientific research and “point-of-care” testing in medicine.
