A method for the purification of a desired protein from a protein preparation includes conditioning the protein preparation by treatment with soluble organic multivalent ions, immobilized organic multivalent ions, or both, optionally in the presence of supersaturated allantoin, thereby removing at least 90% of chromatin, then (1) precipitating the desired protein with a nonionic organic polymer in the presence of non-protein-precipitating salts at greater than physiological concentration to provide a precipitate of the desired protein; or (2) precipitating the desired protein with a nonionic organic polymer in the absence of non-precipitating salts at greater than physiological concentration to provide a precipitate and subsequently washing the precipitate with a nonionic organic polymer in the presence of non-protein-precipitating salts at greater than physiological concentration.

The invention provides methods and apparatus for the selective isolation of phosphorylated and glycosylated proteins and their fragments. A lanthanide metal cation is used to precipitate proteins or protein fragments containing phospho groups and/or glyco groups. The sample preparation method can be used for many types of biological samples, including HeLa cells, food, and human cerebrospinal fluid. The proteins are isolated, recovered and ready for analysis by mass spectrometry or other analytical methods allowing detection limits down to the femtomole level. The method and apparatus are valuable tools in the field of protein analysis and diagnostics.

This invention relates to the separation of complex protein mixtures by lowering (desalting) or by raising (salting-out) their electrolytic (ionic) concentration by electrodialysis followed by chilling, pH adjustment, filtration, and/or centrifugation and optionally thereafter restoration of the lost electrolyte or removal of the gained electrolyte and water. This technique is especially adaptable for therapeutic plasma exchange performed in situ for immunepheresis i.e. removal of globulin or immunecomplexes implicated in autoimmune diseases. The techniques are also applicable for antihemophilic factor separation and purification, and separation of proteins from whey.

A method of co-extracting multiple proteins from chicken egg white is provided. Precipitate proteins step-by-step with polyethylene glycol, then separate the proteins via Q Sepharose Fast Flow anion-exchange chromatography. The method of co-extracting multiple proteins from chicken egg white is capable of obtaining five proteins simultaneously in one extraction process, and the protein products have high purity and high recovery ratio. And the method can be carried out only requiring common-used chromatographic filler, greatly reduces production cost, and provides favorable factors for large-scale industrial production of extracting protein from egg white.

A method and kit for precipitating protein from an aqueous solution consists of a quaternary ammonium cationic surfactant and a chaotropic salt, used in combination to reversibly precipitate proteins from the aqueous solution. Precipitation separates proteins that are entrapped, non-specifically, in the quaternary ammonium cationic surfactant:chaotropic salt complex as a pellet. The precipitated protein pellet is purified further by washing with an organic solvent to remove the surfactant:chaotrope complex, leaving behind pure, concentrated proteins that can be washed to purify further the protein and/or re-dissolved in whatever buffer is most suited for any successive procedure, such as analysis.
