Abstract: The present invention discloses a process for manufacturing a formulation comprising a drug substance, said drug substance comprising a recombinant Listeria comprising a human papilloma virus (HPV) antigen fused to a Listeriolysin O (LLO) protein fragment. The invention further discloses methods of using treating, protecting against, and inducing an immune response against cervical cancer comprising administration of the recombinant Listeria strain. Abstract: In alternative embodiments, provided herein are transcription/translation (TX-TL) systems and methods of using them for use as rapid prototyping platforms for the synthesis, modification and identification of natural products (NPs), and natural product analogs (NPAs) and secondary metabolites, from biosynthetic gene cluster pipelines. In alternative embodiments, exemplary TX-TL systems as provided herein are used for the combinatorial biosynthesis of natural products (NPs), natural product analogs (NPAs) and secondary metabolites. In alternative embodiments, exemplary TX-TL systems as provided herein are used for the rapid prototyping of complex biosynthetic pathways as a way to rapidly assess combinatorial and biosynthetic designs before moving to cellular hosts. In alternative embodiments, these exemplary TX-TL systems are multiplexed for high-throughput (HT) automation and for prototyping engineered platforms for the synthesis or modification of natural products (NPs), and natural product analogs (NPAs) and secondary metabolites analogs.

Abstract: A method of preparing a library of small interfering RNA (siRNA) expression systems for producing siRNA for silencing of target genes by inducing degradation of target gene RNA expression products includes: (i) isolating RNA of one or more target genes from a cell population; (ii) generating RNA fragments from the isolated RNA; (iii) converting the RNA fragments into dsDNA fragments; and (iv) cloning the dsDNA fragments into vectors for forming cloned vectors, each vector including one or more promoters and at least one restriction enzyme site capable of accepting the insertion of at least one dsDNA fragment such that siRNA can be produced. Methods for producing siRNA from the siRNA expression system and methods of identifying a functional target gene for treatment by using the siRNA produced from the siRNA expression system and for identifying RNAi therapeutics are also provided.
