Abstract: Provided is a method for producing an artificial recombinant virus of the family Reoviridae, the method comprising the steps of:
(1) introducing a FAST protein expression vector and/or a capping enzyme expression vector into host cells;
(2) introducing a vector containing expression cassettes for individual RNA genome segments of a virus or introducing a set of single-stranded RNA transcripts from the expression cassettes into host cells; and
(3) culturing the host cells.
The method of the present invention allows more efficient production of an artificial recombinant virus of the family Reoviridae as compared with conventional methods and allows artificial recombinant rotavirus production without using a helper virus.-

Abstract: In related-art methods of differentiating pluripotent stem cells into a desired cell type, there has not been established a differentiation induction method using human ES/iPS cells and being highly efficient. Many attempts have been made, including a stepwise differentiation induction method based on the control of culture conditions or the addition of, for example, various cell growth factors/differentiation factors to a culture solution, but the use of complicated culture steps is a big problem. A method of inducing differentiation into a desired cell type within a short period of time and with high efficiency by use of a pluripotent stem cell that actively undergoes cell differentiation, which is obtained by reducing an undifferentiated state of the pluripotent stem cell, has been developed, and thus the present invention has been completed.

Abstract: The purpose of the present invention is to provide an agent for restoring a visual function or an agent for preventing the deterioration in a visual function, which has an excellent visual function restoring ability. The agent for regenerating a visual function or the agent for preventing the deterioration in a visual function according to the present invention contains, as an active ingredient, a chimeric protein having both an amino acid sequence for a microorganism-origin ion-transporting receptor rhodopsin and an amino acid sequence for an animal-origin G-protein-coupled receptor rhodopsin. The chimeric protein is preferably one in which an amino acid sequence for a cytoplasm-side second loop and/or a cytoplasm-side third loop in the amino acid sequence for the microorganism-origin ion-transporting receptor rhodopsin is substituted by an amino acid sequence for a cytoplasm-side second loop and/or a cytoplasm-side third loop in the G-protein-coupled receptor rhodopsin.

The present invention provides a method for inducing pluripotent stem cells to differentiate into retinal ganglion cells that can be used for clinical application. Such method is a method for producing retinal ganglion cells with elongated axons comprising the following steps: (a) a step of inducing pluripotent stem cells to differentiate into retinal progenitor cells via floating culture; (b) a step of inducing the retinal progenitor cells obtained in step (a) to differentiate into retinal ganglion cells via floating culture; and (c) a step of allowing axons to elongate via adhesion culture of the retinal ganglion cells obtained in step (b).
