摘要: A method for producing eyefield progenitor cells, including: (a) obtaining a starting population comprising human pluripotent stem cells (hPSCs) that are dissociated to essentially single cells; (b) culturing said hPSCs to a contact-inhibited monolayer; (c) contacting said hPSC monolayer in a primary differentiation medium to generate a homogeneous, contact-inhibited monolayer of anterior neuroectodermal cells (ANECs); (d) dissociating said homogeneous ANECs from (c) into essentially single cells; (e) forming dissociated ANECs into size-controlled and homogeneous 3D aggregates (ANEBs), wherein the ANEBs are 3D aggregates of anterior neuroectodermal cells that are distinct from embryoid bodies; and (f) culturing said ANEBs in a primary differentiation medium in suspension to further differentiate them to Eyefield Progenitor Cells (EFPCs).

摘要: The present disclosure relates to the use of laminin-521 in obtaining retinal pigment epithelium (RPE) cells. Pluripotent human embryonic stem cells are cultured on plates coated with recombinant laminin-521 (laminin-11), in totally defined and xeno-free conditions. A first cell culture medium contains a growth factor, and a second cell culture medium does not contain growth factor. The stem cells are first exposed to the first cell culture medium, then exposed to the second cell culture medium for a longer time period. After a number of weeks, clinical grade RPE cells are obtained from the stem cells.

摘要: There is provided a polymeric microsphere comprising a thermally responsive monomer crosslinked with a functional group monomer, wherein the functional group monomer comprises at least one of a carboxylic acid functional group or an amine functional group. The thermally responsive monomer is preferably N-isopropylacrylamide (NIPAM), and the microspheres preferably comprise a coating of polymerized catecholamines (e.g. DOPA). There is also provided a method of preparing the polymeric microsphere and uses of the polymeric microsphere in culturing, harvesting, or expanding stem cells or stromal cells. Preferably, the cells, e.g. hMSCs (human mesenchymal stem/stromal cells), are expanded or harvested in serum-free and xeno-free medium.

摘要: A method for stably amplifying a pluripotent stem cell comprises the following steps: (a) a cell implantation step: implanting pluripotent stem cells directly into a porous scaffold such that the porous scaffold contains 1×104 or more of the pluripotent stem cells; and (b) a cell amplification step: immersing the porous scaffold in a specific culture medium which is xeno-free (XF) and performing amplification culture at an ambient temperature of 35.5-39.5° C. and a CO2 concentration of 5% to obtain the amplified pluripotent stem cells, wherein the amplified pluripotent stem cells aggregate to present an embryoid body state. The amplification method of the present disclosure can easily obtain an excellent effect of increasing an amplification multiple of the pluripotent stem cells to about 3 times or more.

摘要: Methods for generating human arterial endothelial cells under defined conditions in the absence of insulin are described. In particular, provided herein are efficient, defined, and scalable methods for generating human arterial endothelial cells from human pluripotent stem cells. Also provided herein are uses of human arterial endothelial cells obtained according to these methods. For example, methods of treating peripheral arterial disease and methods of screening agents for that effect adhesion of leukocytes to arterial endothelial cells are also provided.

摘要: Methods for generating serum-free and/or xenogen-free cardiac explant-derived stem cells (EDC) are provided. These methods may include providing an initial cardiac explant, which has been minced and digested; plating the initial cardiac explant; culturing the plated cardic explant in serum-free and xenogen-free medium; harvesting EDC cells surrounding or emerging from the plated cardiac explant; and optionally performing static expansion of harvested EDC cells in serum-free and xenogen-free media. Serum-free and/or xenogen-free cardiac EDC cells produced by these methods, as well as methods and uses thereof for the treatment of heart failure in a subject in need thereof, are also provided.

摘要: Methods and compositions relating to the production of induced pluripotent stem cells (iPS cells) are disclosed. For example, induced pluripotent stem cells may be generated from peripheral blood cells, such as human blood progenitor cells, using episomal reprogramming and feeder-free or xeno-free conditions. In certain embodiments, the invention provides novel methods for improving overall reprogramming efficiency with low number of blood progenitor cells.
