Abstract:
An object of the present invention is to construct an mRNA which specifically responds to a short RNA sequence and can activate, repress, and regulate the translation of the desired gene, and to construct an artificial cell model system using a liposome comprising the mRNA and a cell-free translational system encapsulated therein. The present invention provides: an mRNA comprising a target RNA-binding site located immediately 5′ to the ribosome-binding site, and a nucleotide sequence located 5′ to the target RNA-binding site, the nucleotide sequence being complementary to the ribosome-binding site; an mRNA comprising a small RNA-binding site located 3′ to the start codon, and a nucleotide sequence located 3′ to the small RNA-binding site, the nucleotide sequence encoding a protein; and a liposome comprising any of these mRNAs encapsulated therein.

It is intended to provide a polynucleotide comprising a viral base sequence, the viral base sequence containing: a first base sequence encoding a viral replication protein, and a second base sequence encoding a viral movement protein, the second base sequence being located downstream of the first base sequence and having a linking site for linking with an exogenous base sequence encoding a polypeptide to be expressed, the linking site being located downstream of the second base sequence, the second base sequence being obtained by modifying with a base sequence in a native sequence derived from a virus by insertion, substitution, or addition. By using this, a vector containing a viral base sequence is constructed, and a protein is efficiently produced without worsening growth of a host cell containing the vector.

A guide wire includes a distal core member made of a ferrous alloy which has shape memory properties and superelasticity. The ferrous alloy preferably includes substantially two phases, and has a difference of 100° C. or less between an Af point and an Ms point in a thermal hysteresis of martensitic transformation and reverse transformation. The guide wire may include a proximal core member made of an iron-containing alloy and having a higher modulus of elasticity than the distal core member. The two core members may be joined together by welding to form a core of the guide wire.

A method for the fabrication of nonpolar indium gallium nitride (InGaN) films as well as nonpolar InGaN-containing device structures using metalorganic chemical vapor deposition (MOVCD). The method is used to fabricate nonpolar InGaN/GaN violet and near-ultraviolet light emitting diodes and laser diodes.

Provided is a bilayer membrane vesicle capable of undergoing a phase transition. The bilayer membrane vesicle includes: (a) a fatty acid salt having 6 to 20 carbon atoms; (b) an alcohol or an amine compound having an aliphatic chain of 6 to 20 carbon atoms; and (c) an artificial synthetic lipid or a phospholipid capable of forming a bilayer membrane. Preferably, this bilayer membrane vesicle further contains (d) a tertiary amine as a component of the membrane. Also provided is a method of inducing a phase transition of a bilayer membrane vesicle, the method including the step of adding a dehydrating condensing agent or a dehydrating condensing agent precursor having the property of accumulating at an interface to the bilayer membrane vesicle. By causing the lipids that form a molecular aggregate to chemically change, it is possible to change the physical property and the morphology of the molecular aggregate and control the timing of phase transitions such as membrane fusion. In the membrane fusion, for example, fusion can occur without leakage of the contents of the bilayer membrane vesicle.

Provided is a 3-dimensional confocal microscopy apparatus which is manufactured by combining a confocal microscope and an optical tweezers technique, wherein a pair of lenses for focal plane displacement where one lens is movable in the optical axis direction is arranged between a fixed objective lens and a fluorescent light imaging camera, and the 3-dimensional confocal microscopy apparatus also includes a mean which corrects the aberration of a fluorescent confocal image obtained by the fluorescent imaging camera. Accordingly, it is possible to provide a 3-dimensional confocal microscopy apparatus which can acquire a 3-dimensional image of a specimen during a manipulation of the specimen using optical tweezers without affecting an optical trap.

An expression vector is constructed by transferring recombinant tomato mosaic virus (ToMV) cDNA, in which a coat protein gene of ToMV having a suppressor against a virus resistant reaction has been substituted by a GFP gene, into the downstream of a promoter capable of inducing steroid hormone-dependent transcription. In a transformed tobacco BY-2 cell obtained by transferring the above expression vector into a tobacco BY-2 cells, steroid hormone-dependent transcription is induced, thereby enabling the amplification of mRNA of the GFP gene and induction of the expression of GFP.

A Josephson quantum computing device and an integrated circuit using Josephson quantum computing devices which can realize a NOT gate operation controlled with 2 bits will be provided. The Josephson quantum computing device (1) comprises: a superconducting ring member (10) having a π-junction (6) and a 0-junction (7); and a quantum state detecting member (20) constituted by a superconducting quantum interference device arranged outside of the superconducting ring member, wherein a bonding and an antibonding state brought about by a tunneling effect between a |↑> and a |↓> state as two states degenerate in energy of the superconducting ring member (10) are regarded as quantum bits. The bonding and antibonding states as the quantum bits are read out by the quantum state detecting member (20). The two bit controlled NOT gate operation can be performed by the two quantum bits comprising said quantum bits.

Long-lived charge separation is attained with high efficiency by using a supramolecular complex utilizing pi-pi interaction. A supramolecular complex is formed by pi-pi interaction between extended viologen, the extended viologen having heteroaryls coupled together by a linking group with pi-electron conjugated system, and porphyrin. As a result of study of photoelectron transfer reaction, it has been found that within the supramolecular complex, efficient electron transfer occurs from the singlet excited state of porphyrin to the extended viologen, thereby obtaining a charge separation condition of extremely long lifetime exceeding 1 millisecond.
