摘要: A method of barcoding is provided. The method comprises: (a) obtaining a population of cells or cell organelles in a first volume, wherein the cells or cell organelles comprise target molecules that are associated with a first assayable oligonucleotide subunit; (b) separating the cells or cell organelles into a plurality of second volumes, wherein at least some of second volumes receive a single cell or cell organelle from the population of cells or cell organelles; and (c) associating a plurality of second assayable oligonucleotide subunits with the first assayable oligonucleotide subunit in the second volumes, wherein at least some of the second volumes each receive a different second assayable oligonucleotide subunit, to produce at least some nucleic acid molecules that comprise a first assayable oligonucleotide subunit and a second assayable oligonucleotide subunit.

摘要: The present invention discloses a nucleic acid extraction apparatus and a method using the apparatus for extracting nucleic acids and amplifying target nucleic acids. The nucleic acid extraction apparatus comprising: a nucleic acid extraction element, a waste storage chamber, and a reaction chamber, wherein the reaction chamber is selectively in communication with the nucleic acid extraction element and the waste storage chamber, and the apparatus realizes fluid exchange through the pressure between the waste storage chamber and the reaction chamber. In the present invention, by using an integrated control system, the conventional manual process of nucleic acid extraction and purification is integrated into a fully automatic closed process, making the operation more conveniently and quickly and improving the efficiency of experimental work.

The transposome can comprise a transposase (e.g., Tn5 transposase), a first adaptor having a first 5′ overhang, and a second adaptor having a second 5′ overhang. The methods can comprise contacting a permeabilized cell comprising a nuclear target associated with dsDNA, such as genomic DNA (gDNA), with the compositions provided herein to generate a plurality of nuclear target-associated dsDNA fragments (e.g., nuclear target-associated gDNA fragments) each comprising the one or two single-stranded overhangs.

摘要: Methods of specifically capturing and processing nucleic acids from samples in a simplified, streamlined workflow. Media conditions and/or workflow improvements provided herein permit workflow steps such as cell lysis, nucleic acid denaturation, and target nucleic acid capture and immobilization to be performed in a “one-pot” reaction with minimal or no subsequent washing or rinsing steps prior to enzymatic processing. The media conditions include particular salt and/or detergent concentrations, and the workflow improvements include improved purification steps. The media conditions and workflow improvements effectively isolate target nucleic acid for downstream enzymatic processing reactions while avoiding levels of contaminants that inhibit such reactions.
